Objective:Different grade gliomas specimen,that collected in clinical and stashed in the refrigerator with- 80℃,get some tissue from the gliomas specimen and make it as paraffin section,to observe the DNA methylation case of MGMT in the gliomas specimen,besides, get same tissue from the gliomas specimen then using the Tissue DNA extraction kits to get the whole-genome DNA,using the methods of methylation nest PCR and DHPLC to test and verify the DNA methylation case of MGMT in the gliomas specimen. Methods:In part I,get some tissue from the human brain glioma specimen that we have collected,and the normal tissue near by the glioma.using the immunohistochemistry to test it,then to observe the MGMT DNA methylation both of the glioma tissue and the tissue near by the glioma.In part II, making the human brain glioma to be a tissue specimen,then using the methods of methylation nest PCR and DHPLC to validate the DNA methylation case of MGMT in human brain glioma. Result:1.Based on different grade brain glioma,according to WHO,we found that there are none of the eleven WHO I glioma exist the MGMT DNA methylation.In the thirty cases of two WHO II glioma,we found there are teen of it exist the MGMT DNA methylation,the percentage is 46.7%(14/30). In the eleven cases of two WHO II–III glioma,we found there are two of it exist the MGMT DNA methylation,the percentage is 36.4%(4/11). In the twenty-two cases of two WHO III glioma,we found there are nine of it exist the MGMT DNA methylation,the percentage is 59.1%(13/22). In the twenty-six cases of two WHO IV glioma,we found there are five of it exist the MGMT DNA methylation,the percentage is 30.8%(8/26).2. Meanwhile,we used the methods of methylation nest PCR and DHPLC to test the MGMT DNA methylation,and we used the immunohistochemistry to observe the MGMT DNA methylation of the gliomas specimen,we foung the results are the same with above. Conclusion:By using the methods of the methylation nest PCR and the DHPLC and the immunohistochemistry to test different grade human brain glioma, whether it exists the MGMT DNA methylation or not,we found that in the low grade brain glioma,the frequency of occurrence about MGMT DNA methylation is more frequently than in the relatively high grade human brain glioma.But,no matter the the low grade brain glioma or in the relatively high grade human brain glioma,it all exists the case of the MGMT DNA methylation.so,it indicates that the MGMT DNA methylation is one of the factors lead to the human brain glioma occurrence.
