Objective: By studying the effects of Banxia Xiexin Decoction medicated serum on inhibition of gastric cancer cells(BGC-823) in fluids to injury the(HMrsv5) cells-a human peritoneal mesothelial cell line, to explore the effect of Banxia Xiexin Decoction medicated serum intervention in gastric cancer peritoneal metastasis and its related mechanism. For the Banxia Xiexin Decoction medicated serum inhibition of gastric cancer cells in peritoneal mesothelium cell damage to provide experimental basis.Methods: Collect gastric cancer cells(BGC-823) the secretion of supernatant fluid peritoneal mesothelium cell(HMrsv5) with human trained in the normal rabbit serum with100ul/m L、200ul/m L、400ul/m L concentration of Banxia Xiexin Decoction medicated rabbit serum to intervene. Experiments are grouped into control group(normal) peritoneal mesothlium cell, cells in the intervention group(contact of peritoneal mesothelium cell) on gastric cancer, Banxia Xiexin Decoction drug serum1, 2, 3 groups after the intervention.(1)were observed under The light microscope each group of cells morphology and cells structure changes;(2) MTT method was used to detect the growth vitality of each group of cells line;(3)flow cytometry instrument and fluorescent dye were used to test each group of cells proliferation apoptosis changed;(4)To detection of different cell group on the oxidative stress index of SOD, MDA, GSH levels;(5) The Western Blot method was used to detect different cell group the Bcl-2, Bax, TGF-β1 the change of protein content;(6) ELISA method was used to detect each group of cells supernatant of TGF-β1protein expression.Results:(1)As Banxia Xiexin Decoction medicated serum concentration of 2 times increase, contact with gastric cancer cells in fluids and the peritoneal mesothelial cell form by loose slenger fibrous gradually became roung are arranged closely, cell skeleton by tiny microfilament progressively more bare area gradually disappear;(2)Fluorescent staining showed contact with gastric cancer cells in fluids and the peritoneal mesothelial cell the color is orange red more nuclear and cracking phenomena,As Banxia Xiexin Decoction medicated serum concentration increase as cell apoptosis rate gradually decreases;(3)Contact with gastric cancer cells in fluids and the peritoneal mesothelial celldetermined by MTT reduction ability is lower than other groups, as medicated serum concentration increase as the peritoneal mesothelial cell reduction ability increases gradually;(4)The MDA level is lowest, SOD and GSH level is the highest in control group, MDA level from high to low in turn in the 100μl/ml、200μl/ml、400μl/ml Banxia Xiexin Decoction medicated serum but all is lower than the model group, SOD and GSH levels were higher than the model group;(5)Contact with gastric cancer cells in fluids and the peritoneal mesothelial cell compared with control group that G0/G1/G2/M ratio rose to 53.5% from 79.3%, S phase proportion reduced from 46.5% to 46.5%, medicated serum concentration gradually increased compared with model group that G0/G1/G2/M ratio gradually reduce S phase proportion increased;(6)Banxia Xiexin Decoction medicated serum can inhibition of gastric cancer cells the secretion of supernatant fluid to promote peritoneal mesothelium cell with human on the high expression of Bax protein and can raise the Bcl-2 protein, suppress the secretion of TGF-β1protein;(7)ELISA method was used to detect each group of cells supernatant of TGF-β1protein expression, contact with gastric cancer cells in fluids and the peritoneal mesothelial cell compared with control group TGF-β1protein expression increased but TGF-β1protein expression have been gradually decreasing in the 100μl/ml 、 200μl/ml 、 400μl/ml Banxia Xiexin Decoction medicated serum.Conclusion: Banxia Xiexin Decoction medicate serum can inhibition of gastric cancer cells the secretion of supernatant fluid to injury the peritoneal mesothelium cell with human,and the concentration dependence of obviously, this mechanism may be concerned with oxidative stress resistance and increase the expression of Bcl-2 and inhibit the expression of Bax and inhibition of TGF-β1 protein secretion.
