Object:This study aimed to observe the effect of Quzhuotongbi prescription on the structure of intestinal flora in hyperuricemia model rats, and speculate the mechanism of Quzhuotongbi prescription reducing the level of uric acid in the perspective of the intestinal flora.Methods:The hyperuricemia rat model was established by feeding with high purine fodder. The normal control group, model control group and Chinese medicine group (low, middle and high dose) were established respectively. The blood was collected from all groups before and after molding at2,3,4 and 5weeks for detecting the level of blood uric acid. The rat feces were collected at the 5th week. The structures of rat intestinal flora were detected and analyze dusing 16s rRNA gene sequencing technology to determine the microbial community structure and abundance.The effect of Quzhuotongbi prescription on the rat intestinal flora and correlation between intestinal flora change and blood uric acid level were statistically analyzed using the principal component analysis (PCA) and redundancy analysis (RDA).Results:The hyperuricemia rat model was successfully established. The 16s rRNA gene sequencing results showed the intestinal flora structures being difference between the normal group and model group. Compared with the normal rats, the rats in the model group had lower Firmicutes, higher Bacteroidets and Spirochaeies. Quzhuotongbi prescription could reduce the levels of blood uric acid, serum Cr, BUN and serum XOD in hyperuricemia model rats. Meanwhile, new flora categories appeared in Quzhuotongbi prescription rats including Chlamydiae and Deferribacteraceae. The higher dose of Quzhuotongbi prescription resulted in increased percentages of new flora categories. The correlation between intestinal bacteria abundance and physiological biochemical indexes showed that abundance of Prevotellawas positively correlated to uric acid concentration and hepatic index in the model group rats, abundance of CF231 was positively correlated with serum xanthine oxidase.Parabacteroides abundance was positively correlated with renal index.In the normal rats,abundance of Blautia was positively correlated with hepatic xanthine oxidase.Conclusion:(1) Quzhuotongbi prescription can significantly reduce the level of serum uric acid in hyperuricemia model rats, and has a proportional relation with dosage.(2) Different doses of Quzhuotongbi prescription have different effects on the species of intestinal flora in rats:Gracilibacteraceae content rose significantly in the rats fed with a low dose of Quzhuotongbi prescription, while there was no obvious difference in the rats fed with medium and high doses of Quzhuotongbi prescription; besides, the dosage of Quzhuotongbi prescription had no significant effect on biodiversity indexes.
