| The Sambucus Williamsii Hance belongs to the genus Sambbucus chinenis,As a species of genus,Sambbucus Williamsii Hance is also called Xu-gu-mu.Their sexual flavors are sweet, bitter, flat. The Sambucus Williamsii Hance are mainly distributed in northeast China, the second, it also have been grown in China, north China, east China, central China, GanSu, SiChuan, YunNan and other places.The Sambucus Williamsii Hance has a long history in medicinal effects and has many pharmacological effects, such as promoting bone healing, anti-inflammatory, antiviral, enhance immunity, etc.In recent years, we has the constantly researched in the pharmacodynamics study of the Sambucus Williamsii Hance, and obtained many important results, but the research of absorption, distribution and excretion is not many in the body, therefore we made the experiment of pharmacokinetic about SD rats in vivo.At the beginning of this experiment,we establish the detection methods of Morroniside in plasma, detection methods of Morroniside in variety of tissue organs, and detection methods of Morroniside in the faeces, urine, and bile. Meanwhile, we make the investigation of methodology for all methods, which conclude strong specificity, high sensitivity, good precision, stability and recovery rate in all methods comply with the conditions of pharmacokinetic study.Using the method,we investigate the pharmacokinetic process of Morroniside in vivo, after different doses of the Sambucus Williamsii Hance by intragastric administration. The result shows that the Morroniside has a rapid absorption and elimination, but a low bioavailability in SD rats after different doses of the Sambucus Williamsii Hance in intragastric administration. We investigate the rule of tissue distribution about Morroniside in SD rats in vivo,when after different doses of the Sambucus Williamsii Hance by intragastric administration.And the results show that the Morroniside has quickly the speed of distribution in rat tissues in the body, higher concentration in the stomach and small intestine in initial stage of dose then slowly lower in later. What’s more, adequate blood flow of others organs reached maximum concentration in 5 minutes, lungs and kidney to reach the highest concentration in 60 minutes. The Morroniside has a lower content in brain tissue, not through the blood brain barrier, and no obvious accumulation phenomena in various organizations. Apart from this, we discovery excretion situation of the Morroniside in urine, bile and excrement. The results show that the urine is the main excretion way of prototype Morroniside drugs, prototype Morroniside drugs are low content in the bile which be not detected in feces.This experiment provides data support for further study of the Sambucus Williamsii Hance. |
