| Objective: we investigate the association between ADAM33, IL-4R genes polymorphisms and childhood asthma to research pathogenesis, predict and treatment of asthma. Methods: There were 153 children with asthma and 103 controls in this research. We selected 8 SNPs including 6 in ADAM33 gene and 2 in IL-4R gene by reading lots of correlative papers. We combined PCR, Single base extension and matrix-assisted laser desorption/ ionization-time of flight(MALDI-TOF) analysis technologies functioning in Mass ARRAY system to detect the sample genotypes. We evaluated genotypes and alleles in two groups to analyze the association between genes and childhood asthma. Results:(1)There were no significant differences in gender and age of two groups(P>0.05).(2)No significant differences were detected of genotypes or alleles in rs2280090(T2), rs2787094(V4), rs44707(ST+4), rs511898(F+1), rs528557(S2) and rs12927172 sites between two groups(P>0.05).(3) There were no significant differences of alleles frequency in rs574174(ST+7) site between asthma and control groups(P > 0.05). But there was a significantly difference of ST+7 genotypes when TT genotype compared with CC genotype(OR:0.234, 95%CI: 0.060-0.910, P<0.05) and no significant difference when TC genotype compared with CC genotype between two groups(OR:0.975, 95%CI: 0.534-1.78, P > 0.05). Furthermore, compared with TT genotype, there was a significantly difference of CC+TC genotype in two groups(OR: 0.235, 95%CI: 0.061-0.908, P > 0.05).(4) No significant differences were detected of allele frequency of rs2107356 site in asthma and control groups. But there was a significantly difference of rs2107356 genotypes when TC genotype compared with CC genotype(OR:0.548, 95%CI: 0.307-0.979, P<0.05) and no significant difference when TT genotype compared with CC genotype between two groups(OR:0.516, 95%CI: 0.232-1.15,P>0.05). Furthermore, compared with CC genotype, there was a significantly difference of TT+TC genotype in two groups(OR: 0.541, 95%CI: 0.310-0.946, P<0.05). Conclusion:(1) The TT and CC+TC genotypes of rs574174(ST+7) polymorphism of ADAM33 gene may decrease the risk of susceptibility to childhood asthma.(2)The TC and TT+TC genotypes of rs2107356 polymorphism of IL-4R gene also may decrease the risk of susceptibility.(3)There are no association between childhood asthma and the rest of testing loci of ADAM33 and IL-4R genes. |
