Objective:Essential hypertension is a complex disease which can be triggered by heredity or environment or both, as well as a heavy load to the society and family. It’s the main risk factor of cardiovascular diseases. If the blood pressure maintaining a continuous high level, it could damage your body or even worse, take your life. Dai nationality is one of extra minority of Yunnan province. In recently years, The Dai’s prevalence of hypertension has a huge increase than ever before. From 1980’s, our life style and diet structure has been changed. A nutrition research of Dai shows that the salt intake is far more than a normal person daily normal amount, which means they eat too much salt than ever before. Our research mainly studies the methylation of candidate genes which related to sodium metabolism and the relatives between SNP locus and Essential hypertension in Dai nationality.Methods:Our research can be divided into gene ADD1 methylation part and the relevance between SNP polymorphism and essential hypertension part. In The first part, fifty hypertension patient and fifty healthy people were all selected from Dai in Yun Nan province. Their blood samples were all fasting blood which are taken from peripheral vein. Then, Extract DNA from blood sample and by means of Bisulphite Sequencing and pyro-sequencing technology we got nine CpG locus methylation’s status in gene ADD1’s promoter region. After that, we use the status to statistic and got 17 patients in hypertension group and 25 healthy people in normal group. The second part is to establish a genotyping by three SNP locus. Locus rs4762 and rs699 are located in gene AGT, locus rs4524238 is located in gene AGTR1. This part blood samples were all collect from Dai in Dehong and Xishuangbanna. In these samples, hypertension patients were 595 cases, and healthy people were 813cases. We use paramagnetic particle method to extract DNA from peripheral blood, and use Taqman technology to finish the genotyping. Next we use DNA sequencing to test our result. We use student T to analyze the methylation status and chi-square test to analyze the SNP genotyping result.Results:In the research of gene ADD1 shows that from the sixth to the ninth locus has relevance in nine CpG locus of gene ADD1 promoter region in Dai. The hypo methylation of CpG1 locus has statistic difference in the whole sample (P=0.009) and female sample (P=0.018). The CpG6 locus has statistic difference in male sample (P=0.024). The CpG6 locus hypo methylation has statistic difference in whole sample (P=0.047). In the SNP genotyping test, we found that the methylation of locus rs699 has statistic difference in the whole sample (P=0.002) and male sample (P=0.009). However, the locus rs4762 and locus rs452438 only have statistic difference in male sample (P=0.029, P=0.021).Conclusion:Our research shows that the hypomethylation of gene ADD1 has related to essential hypertension from dai in Xishuangbanna and the methylation of gene ADD1 has heterogeneity in different nationalities. The polymorphism of locus rs699 has related to essential hypertension, and the polymorphism of locus rs699, rs4762, rs4524238 has related to male essential hypertension from dai in Dehong and Xishuangbanna.
