Objective: To observe of Xiaochaihu Decoction on functional digestive adverse(FD) of rat liver stagnation and spleen deficiency of brain gut peptide SP, ghrelin, VIP protein expression, from the brain gut axis approach to explore the mechanism of Xiaochaihu Decoction in the treatment of FD.Method: According to the random number table, 60 SD rats(SPF level) were randomly divided into blank group(A) and Model preparation group(M). Group A which have 12 rats(half male and half female) with normal feeding, without any intervention. Group M with complex pathogenesis modeling method were made for21 days. After the end of the modeling, according to the random number table the successful model of 36 rats were randomly assigned to three groups(12 rats in each group, half male and half female), model group(B), Xiaochaihu Decoction group(C),domperidone(D),the rats on group B, were bound 30 min,and given gavage of 0.9%Na Cl solution; the rats on group C and D group, were bound 30 min, and given service Xiaochaihu Decoction solution and domperidone solution for 14 days. The expression of SP, Ghrelin and VIP protein were detected by Western blot(Western-blot) in the hypothalamus, spinal cord, gastric antrum and colon tissue.Results:1. After the end of the modeling rats showed activities reduce, body weakness,irritability, withered hair messy, stool pond when node; weight loss; rat wearing number, stand number and modified number decreased significantly, decreased activity; urinary D-xylose make discharge rate decreased. The difference was statistically significance.2. After the end of treatment, compared with group A, group B rats hypothalamus,spinal cord, gastric, colon tissue VIP protein expression increased significantly, SP and the expression of ghrelin decreased significantly compared with statistical significance(P < 0.05). Compared with group B, C and D in the two groups of rats in the hypothalamus, spinal cord, gastric, colon site VIP protein expression decreased obviously, SP and the expression of ghrelin was significantly higher, the differences were with statistical significance(P < 0.05). But there was no significant difference between the two groups(P>0.05).Conclusion:1. Application of complex etiology modeling method can successfully establish the model of liver stagnation and spleen deficiency FD rats.2.Xiao Chai Hu decoction can be upregulated in the hypothalamus, spinal cord,gastrointestinal tissue ghrelin, SP protein expression and down-regulation of protein expression of VIP in the organization and its role and domperidone group, the regulation of abnormal brain gut peptide to restore intestinal homeostasis in the brain is Xiaochaihu Decoction in the treatment of FD important molecular mechanism.3. The results of this study showed that Xiao Chai Hu decoction can regulate the dynamic balance between brain and intestine by correcting the abnormal brain gut peptide, which can effectively improve the gastrointestinal motility.
