BackgroundPulmonary fibrosis?PF?is a pulmonary interstitial disease characterized by early alveolar inflammation and abnormal proliferation of a large number of fibroblasts?FB?and the deposition of extracellular matrix?ECM?.Its prognostic effect is not ideal due to the lack of effective drug treatment.Silicosis is mainly caused by PF results from the long-term inhalation of dust with high content of free silica?SiO2?,but its formation mechanism has still not clear yet.Rat models with SiO2-induced PF was studied in this research to make a comprehensive analysis of differently expressed proteins between in the rats group with PF,aiming to figure out the key proteins related to PF.The purpose of this paper was to explore and clarify the formation,development mechanism and diagnosis of PF,and to supply new targets for the diagnosis and prevention of PF.ObjectiveTo identify the expression of differentially expressed proteins in the lung tissue of the rat model with SiO2-induced PF by isobaric tags for relative and absolute quantitation?iTRAQ?.MethodsAfter establishing the rat models with SiO2-induced PF,the protein samples of lung tissue were extracted from rats in the model group and control group,respectively.The proteins were separated by liquid chromatography?LC?after iTRAQ,with identification by mass spectrometry and analysis of biological information.ResultsAmong the identified 894 differentially expressed proteins,there were 443 up-regulated proteins,and 451 down-regulated proteins.These differentially expressedproteins are mainly involved in acute inflammatory response,defense response,complement activation,humoral immune response,immune responses,innate immune response,immune globulin-mediated immune response,adaptive immune response.ConclusionThe differently expression proteins lineages of rat models with SiO2-induced PF by quantitative proteomics technique had certain theoretical value of the occurrence and development of PF,and provide new therapeutic strategies for the treatment of PF.
