Objective To identify difference of proteins expression in delayed encephalopathy after carbon monoxide poisoning(DEACMP),DEACMP model in SD rats were established to screen high sensitive and specific and diagnostic significance proteins.This study was expected to provide theoretical basis and reference for the pathological mechanism,prevention,early diagnosis and treatment of DEACMP.Methods DEACMP model in SD rats were established by the static method of infected.Then the model was verified by Morris maze;Pathologic observation was implemented on brain tissue.iTRAQ technology was implemented on searching differential proteins.Results DEACMP model in SD rats was established successfully.The latency time of control group and poisoned group had no statistical significance before infected,and after infected 1d,3d,5d(p>0.05).The latency time was significant longer of poisoned group than that of control group(p<0.05).On histopathological observation,individual hippocampal pyramidal cells were contractible,the nucleus were pycnotic,hyperchromatic and structure was not clear after infected 1d.These pathological characteristics were aggravated,the layer of hippocampal pyramidal cells were attenuated and the number of hippocampal pyramidal cells were decreased after infected 3d.These pathological characteristics were more aggravated after infected 7,21,28 d.There were 179 significant difference proteins were detected by iTRAQ technology between control group and poisoned group.Among those proteins,the difference of myelin protein lipoprotein,vimentin,calretinin and mitochondrial phosphate carrier protein were particularly significant.Conclusions This study has detected differential proteins of DEACMP in SD rats using iTRAQ technology.Results displayed that there were significant difference proteins between control group and poisoned group.These proteins were expected to provide theoretical basis and reference for the pathological mechanism,prevention,early diagnosis and treatment of DEACMP.
