Background Pulmonary fibrosis(PF)is a disease induced by a variety of causes,with the highest mortality of respiratory system,and its main clinical features are progressive dyspnea and diffuse interstitial fibrosis.Due to the lack of effective drug therapies,its prognosis is not ideal.Paraquat(PQ)is a herbicide that widely used all over the world,and lung is one of the main targets after paraquat poisoning.It is much easily accumulated in lung once touching;and the following pulmonary edema and acute lung injury may cause pulmonary fibrosis.However,its mechanism of action it is not very clear.Rat models with PQ-induced PF was studied in this study to make a comprehensive analysis of differently expressed proteins between in the rats group with PF,aiming to figure out the key proteins related to PF.The purpose of this paper was to explore and clarify the formation,development mechanism and diagnosis of PF,and to supply new targets for the diagnosis and prevention of PF.Objective To identify the expression of differentially expressed proteins in the lung tissue of the rat model with PQ-induced PF by isobaric tags for relative and absolute quantitation(i TRAQ),aiming to explore the proteins related to the occurrence and development of PF.Methods 20 male rats(180-200g)were randomly divided into two groups: rats in the model group was given to 50mg/kg PQ by intragastric administration,while 50mg/kg physiological saline was given to rats in the control group.Rats were sacrificed after 28 d,and histological changes in the lung of rats were detected by HE staining.The expression of proteins were analyzed by i TRAQ combined with LC-MS/MS,the expression difference was analyzed by NCBI data and Proteome Discoverer Software.Results HE staining showed that the PF grade of rats in the model group was greater than that in the control group(P<0.05).Among the identified 268 differentially expressed proteins,there were 194 up-regulated proteins,and 74 down-regulated proteins.Conclusions In this study,the rat model with PQ-induced PF was successfully establishmented,and it has been demenstrated that i TRAQ-LC-MS/MS technique can effectively used for the study on differentially expressed proteins,and a few proteins associated with PF were screened.It plays a important role in the occurrence and development of PF;and may provide theoretical basis for the treatment and prognosis of PF.
