Investigation On The Expression And Clinical Significance Of Epidermal Growth Factor Receptor Family In Gastric Cancer Patients

BackgroundThe prognosis of gastric cancer is poor,and it is the second major cause of cancer related death.Tumor angiogenesis is a key point in the development of solid tumors.Therefore,it is a hot spot to regulate angiogenesis.Epidermal growth factor receptor is a tyrosine kinase family,which plays an important role in cell proliferation,apoptosis,angiogenesis and metastasis.At present,there are many studies on HER family members in gastric cancer,and it has not been reported that the expression of multiple receptor proteins in gastric cancer tissues and the correlation analysis of clinical pathological parameters are not reported.ObjectiveIn this study,the expression of HER family in tissue samples of 164 patients with gastric cancer was detected,to investigate the correlation between the expression of HER(Human Epidermal Growth Factor Receptor)protein and the diagnosis as well as prognosis of gastric cancer.It is expected to provide an important reference for the detection,treatment and prognosis of gastric cancer in the family of the Human Epidermal Growth Factor Receptor.MethodsA total of 164 cases of male patients diagnosed with gastric cancer(120 male and44 female)were analyzed.Using immunohistochemical method and Western Blot to detect expression situation of EGFR,HER-2,HER-3 and her-4,and analysis the correlation of different expression levels and clinicopathological characteristics and prognosis.To analyze the experimental data by the SPSS22.0 statistical software,Measurement data using t test,count data were compared using the chi square,Theprognosis by Kaplan Meier survival analysis and selection of P< 0.05 for differences with statistical significance.ResultsThe results indicated that the positive expression of EGFR in gastric carcinoma was 40.24%(66/164),the positive expression rate of HER-2 was 33.54%(55/164),the positive expression rate of HER-3 was 67.07%(110/164),the positive expression rate of HER-4 was 51.83%(85/164),the positive rate of expression was associated with TNM stage,Pathologic type,depth of invasion,but not gender,age,differentiation degree,tumor location and regardless of the size.Except HER-4,the remaining three were correlated with lymph node metastasis of gastric carcinoma.Western Blot results showed that the expression of HER in gastric carcinoma were significantly higher than that in adjacent tissues.EGFR negative and positive median DFS were 9.34,7.24 months,median OS were 12.43,9.38 months;HER-2 negative and positive group median DFS were 9.28,7.56 months,the median OS were12.69,9.78 months;HER-3 negative group and positive group the median DFS were10.21,7.52 months,median OS were 13.52,10.26 months;HER-4 negative and positive group the median DFS were 10.58,8.22 months,the median OS were14.27,11.09 months.ConclusionsThe positive expression of HER family in gastric cancer tissues exist different degree,they are main factors of poor prognosis.The positive rate of HER-3 was significantly higher than the other three,it’s closely related to the metastasis of liver,it can be a potential biological marker to evaluate the prognosis of gastric cancer,and may be an important target in the future.
