Objective:A metabonomic strategy based on Kidney-Yang Deficiency mice model infected with influenza virus. Take medicine disproof, which would contribute to the exploration on the essence of Kidney-Yang Deficiency mice model infected with influenza virus from the perspective of metabonomic. And reaserching the approach and function link intervention of MXF for Kidney-Yang Deficiency mice model infected with influenza virus.Methods: The mices were injected with estriol benzoate for 15 days to cause Kidney-Yang Deficiency, and then infected with influenza virus by nasal. This subject determined the metabolism characters of serum by HPLC-TOF / MS. LC-QE/MS/MS was used to analyze the lipid compounds in the lung of mices. XCMS software was used for data pre-process, and SIMCA-P 11.5 software was used for data analysis. The difference variable between lung tissue and serum-derived in serum of mices; based on pharmacological effects of MXF, and then use HPLC-TOF / MS analysis instrument to explore the main chemical components in MXF and pharmacodynamics substances, and explore the intervention mechanism of MXF intervention.Results: 1. By body weight, rectal temperature, loco motor activity time and the swimming time judge the model copied success or not. Then by analysis the pharmacological indicators derived from traditional Chinese medicine MXF inferred that the MXF established a good inventation effect in the “Kidney –Yang Deficiency” with Exterior syndrome.2. This subject determined the metabolism characters of serum by HPLC-TOF / MS. Significant metabolic disorders were found many biomarkers were identified in ‘Kidney-Yang Deficiency mice model infected with influenza viru’. The plasma level of Leucine, L-Lactic acid, Sphinganine, Lyso PC(18:0), 5-HETE, Linoleic acid in model mice were significantly changed. Which mainly involved in hypothalamus-pituitary-target gland axis, sphingolipid metabolism, metabolism of linoleic and arachidonic and so on. 3. The lipid reaserch indicated that there are two kinds of biomarkers of lipid. Including PC(18:1(9Z)/0:0), DG(14:0/22:0/0:0), Lyso PE(22:0/0:0), TG(15:0/15:0/20:2n6), TG(16:0/16:0/20:2n6), TG(15:0/16:0/18:1(11Z)) and so on. Which has major link with metabolism of linoleic and energy. MXF has obviously function of adjustment of lipid. 4. Using HPLC-TOF/MS to analysis 52 compounds and inferring the possible structures of them, which are 42 alkaloid components and 11 compounds structure were determined. According to the precise molecular weight and fragment information infered 41 components.Conclusion: Traditional Chinese medicine MXF has good effect on “Kidney-Yang Deficiency mice model infected with influenza virus” with Exterior syndrome. There are a variety of Significant metabolic disorders in serum of mices appears callback trend after the drug intervention. The reaserch indicated that MXF has good effect on hypothalamus-pituitary-target gland axis, sphingolipid metabolism, metabolism of linoleic and arachidonic and so on. Lipid is a kind of important biomarker of the model “Kidney-Yang Deficiency mice model infected with influenza virus”. MXF can adjustment them by the way of energy consumption. The main substances of MXF is alkaloid. It highlights that metabonomics is a promising tool for studying the essence of Kidney-Yang Deficiency mice model infected with influenza viru’. Based on the method of metabonomics and start from the point of view the network of the metabolic, we can explorethe essence of the intervention of MXF. Which conform to the principle of holism in TCM and has unique advantage in revealing the essence of "syndrome" and the mechanism of TCM.
