| Background:The purpose of the study is to compare the spectral differences between the tissues of early stage and advanced stage in nasopharyngeal carcinoma(NPC), and assess the non-invasive diagnosis of NPC at different stages using Raman spectroscopy(RS) method.Method:RS measurements were performed to acquire high quality RS spectra on two groups of tissue samples: one group consists of 30 NPC patients at the early stage(T1-2), and the other group was 30 NPC patients at the advanced stage(T3-4). Effective diagnostic algorithms for distinguishing RS spectra between the patients of the early and advanced stages were applied by utilizing Principle components analysis(PCA) and linear discriminant analysis(LDA)Results:During the study, significant differences in Raman spectra were detected between the early and the advanced nasopharyngeal tissues. In both types of tissue samples,spectral peaks can be detected at following sites, 853,937,1004,1032,1094,1209,1268,1304,1340,1450,1527,1660cm-1.In particular, the band intensities located at 1032, and 1209cm-1 were significantly higher in the advanced tissues than those in the early tissues(p<0.05),while the band intensities located at 853,937,1004, 1094,1268,1304,1340,1450,1527,1660cm-1were significantly lower(p<0.05) in the advanced tissues. The PCA-LDA algorithms based on Raman spectroscopic data yielded diagnostic sensitivity of 70%, specificity of 76% and accuracy of 73.3%, respectively.Conclusion:Raman spectroscopy(RS) method combined with the PCA-LDA algorithms has good capability of distinguishing the early and the advanced nasopharyngeal tumor tissues, andit has tremendous potential for the non-invasive detection of early and advanced stage in NPC. |
