| Objective: In this study,we investigated the effects of compound Xiancao granule on urinary protein excretion,creatinine level,changes of renal pathology and Fox O4 transcription factors in diabetic nephropathy rats,to explore the molecular mechanism of compound Xiancao granule on DN ratsMethods: A total of 60 male SD rats,10 rats were randomly divided into normal control group,the remaining rats were used to make the DN model,removing death,modeling unsuccessful and excess rats,the rats were randomly divided into model group(group B),compound Xiancao granule group(group C),irbesartan group(group D),10 rats in each group.Establishment of diabetic nephropathy model induced by high glucose and high fat diet after resection of rat left kidney and injection of streptozotocin(STZ),Compound Xiancao granule group was given 6ml/kg/d compound Xiancao granule solution,The irbesartan group was administered with irbesartan solution at a dose of 12 mg/kg/d,The other two groups were given6ml/kg/d saline,Each group was fed for 12 weeks.And then detected urinary protein,creatinine,urea nitrogen and blood lipid in each group,observation of Pathological Changes of Rat Kidney by HE Staining,western blot and immunohistochemistry were used to detect the expression of Fox O4 protein.Results: 1.Compared with the A group,the levels of urinary protein,creatinine,blood lipid and urea nitrogen in B group were significantly increased(P <0.05).Compared with the B,the above indexes of compound C group and D group were significantly decreased(P <0.05).2.HE staining results showed that the A group of rats with normal renal tissue structure,Dgroup glomerular enlargement,basement membrane thickening,mesangial matrix increased and renal tubular abnormalities,the pathological changes of C group and D group were improved obviously.3.Compared with the A group,the expression of Fox O4 in renal tissue of Bgroup was significantly higher than that of A group.Compared with the B group,the expression of Fox O4 in renal tissue of compound C group was significantly lower than that of B group(P <0.05).Conclusion: Compound Xiancao granule can effectively reduce urinary albumin excretion in DN model rats and decrease creatinine.Improve the pathological changes of renal tissue,and can effectively reduce the expression of Fox O4.Its mechanism may be by reducing the expression of kidney Fox O4,which protect the rat kidney tissue. |
