| In this paper,Caenorhabditis elegans(C.elegans)as a model animal for evaluated the effect about lifespan,heat shock tolerance and antioxidant capacity,and the effect of Maca water extract on body fat accumulation also been assessed.First of all,Maca was broken into power,determined the water extract technology,the the water extract was dissolved and purified and freeze-dried.Effect of water extract of Maca on Physiological Indexes of nematodes had been evaluated.For the lifespan assay,0.5mg/ml and 2mg/ml water extract of Maca water extract had significant differences compared with wild type C.elegans N2,the life expectancy increased 27.3%,the average life expectancy for 1mg/ml Maca blank group was 9.09%.Compared with control group,the average fecundity increased by 12.72% in 2mg/ml Maca high concentration group,and the average fecundity increased 9.79% in 1mg/ml Maca group.For the swallowing rate assay,the treatment groups have little difference with control group,which indicting that the effects of Maca on C.elegans was depended on non-calorie restriction pathway.For the lomaction assay,2mg/ml Maca group had little influence on the movement ability,which is better than control group.The heat shock experiment state was analysised by Log-rank(Mantel-Cox)Test,the survival rate of heat shock group has no significant difference compared with control group.And for the oxidative stress,Maca has good protective effect on 10 m M hydrogen peroxide induced oxidative stress.Otherwise,Maca water extracts have lipid-lowering effect on C.elegans by oil red O staining,which showed that Maca has the potential value for developing as lipid-lowering health care products.In summary,Caenorhabditis elegans was using as a model to evaluate the effect on Maca water extract in this paper,which provide an important experimental basis for further development and utilization of Maca. |
