Objective(1)To study the immunoregulatory effect of different preparations of Mongolian medicine "Chagan Decoction" on the proliferation of mouse spleen lymphocytes in vitro and the immunomodulatory effect of single dose on CTXinduced immunosuppressive mouse model.(2)Compare the different preparation techniques of "Chagan Tang" powder,and screen out the "dry dry powder" freeze-dried powder with better immunomodulatory activity for in-vivo immunomodulatory activity.This thesis is to clarify the effect of Mongolian medicine treatment of the commonly used plague fever classic "Chagan Tang",to play its role in clearing the table and promoting "immature heat maturity",whether to play a role in the treatment of related diseases by regulating the body's immune system.Such as fever and chills,sore throat,headache,cough,chest pain and other symptoms,provide a basis for clinical treatment.Methods(1)By freeze-drying method,spray drying method,direct pulverization method of raw medicinal materials,the medicinal materials of Tumuxiang,Sophora flavescens,Rubus and Hawthorn can be prepared by pulverizing and mixing,respectively."Freeze-dried powder,process powder and ultra-fine powder,and determine the content.(2)An immunosuppressive mouse model was established by a clinically used chemotherapy drug,cyclophosphamide(CTX).Among them,by determining the effect of CTX on non-specific immunity and specific immune function in normal mice,it is judged whether the model establishment is successful.At the level of non-specific immune function,the effect of CTX on the number of peripheral blood cells in normal mice was monitored by blood sampling in the tail vein;the effect of CTX on the phagocytic index of macrophages in normal mice was determined by carbon particle clearance method;The method was used to determine the effect of CTX on the thymus and spleen weight of normal mouse immune organs.At the level of atopic immune function,the effect of CTX on ear swelling caused by delayed type hypersensitivity in normal mice was determined by ear swelling method.(3)The effects of different drug concentrations of "Chagan Tang" lyophilized powder,processpowder and ultrafine powder on the proliferation and immunosuppression of mouse spleen lymphocytes in vitro were studied by CCK-8 cell staining.The effects of a single dose of“Zhagan Tang” lyophilized powder,process powder and ultrafine powder on the immunomodulatory activity of the immunosuppressed mouse model were studied by carbon particle clearance method and gravimetric method.(4)Through the carbon particle clearance method,gravimetric method,CCK-8 cell staining method,ear swelling method,study two ways,preventive administration of "ChaganTang" lyophilized powder and therapeutic administration of"ChaganTang"Effect of dry powder on the immunomodulatory effects of immunosuppressive mouse models induced by CTX.Conclusions(1)Both "Chagan Tang" process powder and lyophilized powder can induce spleen lymphocyte proliferation in vitro in a concentration-dependent manner,and can antagonize the decrease of the number of partial immune cells in the immunosuppressive mouse model induced by CTX.The weight of the immune organ is reduced.(4)"Chagan Tang" can have a certain positive regulation effect on the immunosuppressive mouse model induced by CTX at the level of some immune cells and immune organs,but there is no regulatory effect on the preventive administration of "Chagan Tang".
