| Colon cancer is one of the most common malignant in digest system,with a high incidence and mortality.The treatment of colon cancer has been development greatly,but the prognosis remains undesirable.Therefore,there is still a great need to explore efficacious drugs for colon cancer.Resveratrol(Res)is a kind of natural polyphenols,itprocesses the antioxidant and cardiovascular protection effects,as well as cancer proventing.Res mainly exists in peanut,mulberry,giant knotweed and grapes.As a hot research topic in recent years,the anti-cancer effect of Res has been widely reported,but the detailed molecular mechanism remains unclear still.In this investigation,we determined the anti-cancer activity of Res with CCK8,crystal violet staining,flow cytometry,Anniex V-GFP,PCR and Western blot assays.Theresults showed that Res inhibits proliferation and induced apoptosis inHCT116 cells.Withfurther analysis,we found that Res up-regulates BMP7 either mRNA or protein level in HCT116 cells.Exogenous expression of BMP7 promotes the anti-proliferation effect;On the contrary,BMP7 specific anti-body attenuates the anti-proliferation effect.However,we found that Res exerts no apparent effect on BMPs/Smads signaling,but decreases the level of p-Akt1/2/3 while it was enhanced by exogenous BMP7.At the same time,we found that Res significantly decreases the phosphorylation level of PTEN,and this effect can be enhanced by exogenous BMP7 butattenuated by BMP7 specific anti-body.Our findings suggested that Res may be a potential powerful candidate for colon cancer treatment,and the anti-cancer activity may be mediated by inactivating PI3K/Akt signaling through up-regulating BMP7 to increase the function of PTEN. |
