| ObjectiveTo investigate the mechanism of action on the Shou Tai Wan(STW)decoction which can influence the Kisspeptin-10 in Kiss-1 gene of rat models who suffer from kidney deficiency and LPD(Luteal Phase Defect)abortion.MethodsBy utilizing hydroxyurea and mifepristone,this research set up animal abortional model caused by kidney deficiency and LPD for pregnant SD rats and treat them with water extract of STW decoction and dydrogesterone,in the meantime,a control model group without medication and a normal pregnant group have been established for further comparison.The study collected the morphological change in uterine and ovarian tissues and then measured the rate of abortion,serum progesterone(P)and the content of Kisspeptin-10 and Erk1/2 in the ovary of every group.ResultCompared with the control model group,STW decoction has marked performance in improving the general condition of pregnant rats,lowering the rate of abortion and lifting the progesterone level.According to the H.E staining result of the rats’ uterine and ovary,STW decoction plays a significant role in improving the morphology and function of endometrium and corpus luteum.The immunity fluorescence demonstrated that STW decoction can dramatically increase the expression of Kisspeptin-10(luteinized granulosa cells).The result of RT-PCR and western blot proved the remarkable effect on increasing the expression of kisspeptin-10 and Erk1/2 gene in the ovary and protein level(Such difference has statistical significance compared with control model group.P<0.05).Meanwhile,STW decoction can expedite the protein phosphorylation of Erk1/2 and,as a result,improve its activity.ConclusionBy increasing the expression of Kisspetin-10 and Erk1/2 in ovary,STW decoction may realize its fetus protection mechanism through influencing the change of progesterone and then improve the luteal function. |
