Association Of Serum IL-34 Concentration With Type 2 Diabetes And Obesity

Objective:To explore the association of serum IL-34 concentration with type 2diabetes and obesity.Methods:99 were chosen into two groups according to 1998 year WHO obesity guide,the nonobesity was 32 and the obesity was 67,（the nonobesity arranged from 19.1kg/m² to 24.9 kg/m²,the obesity was equal or larger than 25 kg/m²）.The first group was divided into two subgroups,subgroup 1（n=17）was without T2DM,subgroup 2（n=15）was with T2DM.The second group was also divided into two subgroups,subgroup 3（n=33）was without T2DM,subgroup 4（n=34）was with T2DM.Serum IL-34concentration,hsCRP,fast insulin（FINS）,fast plasma glucose（FPG）,total cholesterol（TC）,triglyceride（TG）,low density lipoprotein cholesterol（LDL-C）,high density lipoprotein cholesterol（HDL-C）,HbA₁c were measured,to determine systolic blood pressure,diastolic blood pressure,waistline circumference（WC）,hipline circumference（HC）,age,sex,calculate HOMA-IR,BMI.Then serum IL-34 concentration was measured using commercial enzyme-linked immunosorbent assay（ELISA）kits according to the manufacturer’s instructions.Results:Data analysis of 99:Serum IL-34 concentration of the subgroup 3 was higher than subgroup 1,2 and 4.The subgroup 4 was also higher than the subgroup 1 and subgroup 2.Subgroup 1 and 2 had no statistical significance.The serum IL-34concentration of subgroup 1,2,3,4 was respectively（167.1±39.38）vs（170.71±35.68）vs（244.48±42.2）vs（211.44±23.83）ng/L.Pearson correlation analysis showed serum IL-34concentration was positively correlated with hsCRP,fast insulin,HOMA-IR,WC,HC,BMI（r=0.672,0.442,0.566,0.678,0.476,0.651,P（27）0.01）.Multiple stepwise regression analysis showed waistline circumference,BMI was independent influencing factors for serum IL-34 concentration（β=0.434,P（27）0.01;β=0.321,P（27）0.01）.After one month of laparoscopic Roux-en-Y gastric bypass（RYGB）surgery of 8（5 women and 3 men）from the group of obesity,hsCRP,BMI,serum IL-34 concentration,waistline was lower,only HOMA-IR had statistical significance.Conclusion:（1）The serum IL-34 concentration is significantly higher during obesity,in obesity group it almost has no relationship with T2DM,so the association between IL-34 and T2DM is unclear.（2）There existed a positive correlation between IL-34 and hsCRP,FINS,HOMA-IR,WC,HC,BMI,and WC and BMI are the independent factors of serum IL-34 level,it may have association with insulin resistance and inflammation,which reveals we can speculate visceral adipose tissue by testing serum IL-34 in order to formulate a corresponding weight loss plan.
