Objective: To observe the influence of vitamin D on ovarian cancer,thus the value of vitamin D in inhibiting the invasion of ovarian cancer by regulating expression of DDX4 can be discribed.Methods: Different concentrations of vitamin D(50ng/ml,100ng/ml,200ng/ml,400ng/ml,800ng/ml)were added into medium for 72 hours and the relative cell numbers were quantified by Cell Counting Kit-8(CCK8).According to the results,proper concertration of vitamin D(100ng/ml)was chosen for the following experiments.SKOV3 cells were transfected with two independent siRNA against DDX4 and relative DDX4 mRNA level was examined by qPCR.According to the results,siDDX4-121 was selected for the following experiments.Western blot and qPCR were used to detect the influence of vitamin D on DDX4 in protein level and mRNA level respectively.Meanwhile,invasion capacity of SKOV3 cells with different treatments was detected by transwell.Results: 1.Invasion ability of SKOV3 cells was inhibited by treatment of vitamin D;2.DDX4 expression in SKOV3 was reduced by treatment of vitamin D;3.Invasion ability of SKOV3 cells was inhibited by down-regulation of DDX4;4.Expression level of DDX4 was positively correlated with the malignancy of ovarian tumor.Conclusion: Vitamin D is able to inhibit the invasion capacity of ovarian cancer cell by down-regulating the expression of DDX4.It is predicted that there may be non-negligible potential value of vitamin D in therapy of ovarian cancer metastasis.
