This paper described the secondary metabolites of two endophytic fungi and the fermentation research of traditional Chinese medicine Cornus officinalis by endophytic fungi.The secondary metabolites produced by plant endophytic fungi have rich biological functions and chemical structure diversity,which are of great significance to discovery novel compounds,study biological diversity,and research new drugs.Microbial fermentation is an important processing method of traditional Chinese medicine,which can transform the components of traditional Chinese medicine,reduce toxicity and increase efficiency,improve the value of traditional Chinese medicine products,and promote the”secondary development"of traditional Chinese medicine.The first chapter reviewed the research progress of genus Fusariunm.The main types of the secondary metabolites and their biological activities were introduced.In chapter 2,the secondary metabolites of F.sambucinum B10.2 were investigated.Fifteen known compounds were isolated and identified as:sambacide(1),12-acetoxy-4,4-dimethylene-24-methylene-5-cholesta-8,14-diene-3,11-diol(2),bisphenol A(3),cyclonerodiol(4),trichoderiol A(5),T-2 tetraol(6),squalene(7),thymidine(8),uridine(9),indazole(10),uracil(11),methyl D-glucoside(12),3-sitosterol(13),eudesmin(14).magnolin(15).Compound 1 was a tetracyclic trnterpene,which had potent inhibitory activity against Escherichia coli,with a MIC value of 32 g/mL.Compound 1 also exhibited the moderate inhibitory activity against acetylcholinesterase,with the inhibitory rate of 47.1%(the final concentration was 50?M).The third chapter focused on the secondary metabolites and their bioactivities of Aspergillus ochralea SX-C7.The potato was fermented with A.ochrateaby solid fermentation.Ten known compounds were isolated from the fermentation extracts and identified as:semivioxanthin(1),6-hydroxy-p-menth-4(5)-en-3-one(2),flavacol(3),magnolin(4),Preussin(5),circumdatins D(6),28-isofucosterol(7),sclerotiamide(8),3?-hydroxyergosta-8,24(28)-dien-7-one(9),3-o-?-D-glucopyranosyl stigmasta-5(6),24(28)-diene(10).The main types of the isolated compounds were alkaloids and sterols.Compound 10 showed obvious inhibitory activity against Bacillus subtilis,the MIC value was 2 g/mL,and the inhibitory effect was at the same grade with positive control.Compound 1 showed potent inhibitory activity against acetylcholinesterase,with an inhibitory rate of 62.3%(the final concentration was 50 uM).In chapter 4,the fermentation modification of C.ornu.s officinali.s with fungi was studied based on the phytochemical research on this herba medicine.C.offi.cinalis was fermented with 50 different strains of endophytic fungi,revealing that Pestalotiopsis negleet TG-3 can facilitate the transformation of some compounds in Cofficinalis.Four compounds,including loganin aglycone(1),3,7-dihydroxy-8-methyloctahydro-4-methyl fornmate(2),sarracenin(3)and 4-epi-alyxialactone(4)were isolated from fermented C.officinalis.Five compounds were isolated from original material C.officinalis,including oleanolic acid(5),?-morroniside(6),loganin(7),(7?)-7-o-methylmorroniside(8)and surose(9).Our research indicated that loganin aglycone(1)and 3,7-dihydroxy-8-methyloctahydro-4-methyl formate(2)were produced under the cooperation of P.neglecta and C.officinalis.This study provides a new idea for the development of C.officinalis and enriches the study of microbial fermentation modification of Chinese medicinal materials.
