| Seaweed species are often regarded as an abundant marine bioresource,which is wildly distributed and have been widely used in food,industrial raw materials,agriculture,animal husbandry production and many other fields.Seaweed fertilizer have been widely used as amendments in crop production systems due to containing rich plant growth-stimulating compounds.Preparation processes and application effects on plants of Sargassum hormeri and Laminaria japonica of seaweed liquid fertilizer were systematically studied.Cellulase,pectinase and protease were applied to prepare seaweed liquid fertilizer.Experimental results were showed as fellow.(1)The conditons for cellulase hydrolysis process were time 8 h,pH 4,temperature 65℃,enzyme addition amount 4%,the extraction rate of alginate of Sargassum horneri and Laminaria japonica were 60.924%and 62.187%respectively;The conditons for pectinase hydrolysis process were time 7 h,pH 3,temperature 55℃ and enzyme additon amount 5%,the extraction rate of alginate ofSargassum horneri and Laminaria japonica were 53.532%and 56.018%respectively;The conditons for protease hydrolysis process were time 7 h,pH 7,temperature 75℃ and enzyme addition amount 4%,the extraction rate of alginate of alginate of Sargassum horneri and Laminaria japonica were 40.740%and 45.813%respectively.(2)Optimum conditions for composite enzymatic hydrolysis in Sargassum horneri were time 6 h,pH 5,temperature 45℃,enzyme addition amount 2%,the extraction rate of alginate was 74.150%.(3)Optimum conditions for composite enzymatic hydrolysis in Laminaria japonica were time 8 h,pH 4,temperature 45℃,enzyme addiion amount 5%,the extraction rate of alginate was78.105%.(4)The ultrasonic extraction conditions of Sargassum horneri and Laminaria japonica were ultrasonic time 25 min,initial temperature 65℃ and ultrasonic power 450W,the extraction rate of alginate were 44.676%and 51.269%respectively.(5)The best hydrolysis method was ultrasonic complex enzymatic treatment,which was enzymatic hydrolysis after ultrasound,the extraction rate of alginate were 80.817%(Sargassum horneri)and 85.994%(Laminaria japonica)respectively.The Strain FSZ39 producing high alginate lyase was screened from the rotten seaweed and enzymatic hydrolysate through the method of preliminary screening with plate and secondary-screening with shake flask.FSZ39 strain were identified by morphological characteristics,physiological and biochemical characteristics and 16S rDNA molecular biological methods,the result shows that FSZ39 strain was identified as Microbacterium paraoxydans.The seaweed liquid fertilizer was fermented by FSZ39 strain,the best process conditions for the preparation of Sargassum horneri liquid fertilizer were as follows:fermentation time was 48 h,the initial pH of fermentation was 7.5,the inoculum volume was 2%,the liquid volume was 100mL·250mL-1 and the fermentation temperature was 30℃.The best process conditions for the preparation of Laminaria japonica liquid fertilizer were as follows:fermentation time was 48 h,the initial pH of fermentation was 7.5,the inoculum volume was 4%,the liquid volume was 100mL·250mL-1 and the fermentation temperature was 30℃.Different concentration dilution of the liquid seaweed fertilizer prepared by enzyme and fermentation method were applied on the seeds and seedlings of brassica chinensis,cucumber and tomato by seed handling and pot experiment.The results showed that:(1)The 600 times diluent of fermented Sargassum horneri and Laminaria japonica liquid fertilizer had a significant effect on the germination of seeds;1000 times of Sargassum horneri liquid fertilizer prepared by enzymes and 400 times dilution of fermented Laminaria japonica liquid fertilizer had a significant effect on all indexes of cucumber seed germination;There were significant differences in all indexes of tomato seeds germination between contrast and 600 times dilution of Sargassum horneri and Laminaria liquid fertilizer prepared by enzymes.(2)The 400 times diluent of fermented Sargassum horneri and Laminaria japonica liquid fertilizer had the most obvious effect on the growth of brassica chinensis seedlings;800 times of Sargassum horneri liquid fertilizer prepared by enzymes and 400 times dilution of fermented Laminaria japonica liquid fertilizer had a significant effect on all indexes of cucumber seedlings;The indexes of tomato seedlings treated by 600 times dilution of fermented were significantly improved.(3)The effect of Sargassum horneri and Laminaria japonica liquid fertilizer on seeds germination of brassica chinensis and cucumber was not obvious,but the effect of Sargassum horneri liquid fertilizer on tomato seeds germination was better than that of Laminaria japonica liquid fertilizer;three kinds of vegetable seed seedlings were evidently enhanced by fermented Sargassum horneri liquid fertilizer. |
