Purpose:The near-infrared laser Raman spectroscopy was used to detect Raman spectra of different pathological types of gastrointestinal polyps,and explore its value and correlation.Methods:Fifty cases of gastric polyps(23 fundic gland polyps,27 hyperplastic polyps)and 78 cases of colonic polyps(49 adenomatous polyps and 29 hyperplastic polyps),21 normal gastric tissues and 21 normal colonic tissues were collected from patients of gastrointestinal polypectomy and gastrointestinal endoscopy in Zhujiang Hospital simultaneously.The tissue samples were detected by 785nm near-infrared laser Raman spectroscopy.The mean Raman spectra of gastrointestinal polyps in different pathological types was analyzed by variance analysis and t-test,PCA-LDA method was used for the differential diagnosis of them,and the ROC curve was used to evaluate the diagnostic efficacy and Spearman correlation analysis of its relevance.The immunohistochemical method was used to detect the expression of ?-catenin protein in gastrointestinal tissues of different pathological types to evaluate whether it was consistent with Raman spectroscopy.Results:The distribution of typical Raman spectrum peaks in gastric and colonic tissues in different pathological types was basically the same,mainly at 9 typical bands of 830cm-1,855cm-1,1003cm-1,1032cm-1,1210cm-1,1323cm-1,1335cm-1,1450cm-1,1655cm-1.1.The Raman peaks of the gastric hyperplastic polyps at the 830cm-1,855cm-1,1003cm'1 bands were lower than normal gastric tissues.The Raman peaks of colonic hyperplastic polyps at the 830cm-1,1003cm-1 were lower than normal colonic tissues,the difference was statistically significant(P<0.05).The Raman peaks of gastric hyperplastic polyps at 1655cm-1,1450cm-1,1335cm-1 1323cm-1 bands were higher and at 855cm-1,830cm-1 were lower than fundic gland polyps.While the Raman peaks of the colonic adenomatous polyps at 1655cm-1,1003cm-1,855cm-1,830cm-1 were higher than colonic hyperplastic polyps,the difference was statistically significant(P<0.05).2.The Raman peaks of gastric hyperplastic polyps at the 1210cm-1,1032cm-1,1003cm-1,830cm-1 were similar to the colonic hyperplastic polyps,and the Raman peaks of fundic gland polyps at the 1032cm-1,855cm-1,830cm-1 were not significant different with colonic adenomatous polyps(P>0.05).The tyrosine was characterized by Raman bands 1032 cm-1 and 830 cm-1,suggesting that it may be related to P-catenin protein.3.The accuracy of PCA-LDA diagnosis of fundic gland polyps,gastric hyperplastic polyps and normal gastric tissues was 93.10%,100%,90.91%,sensitivity was 100%,86.96%,95.24%,specificity was 95.24%,100%,95.92%.The accuracy of PCA-LDA diagnosis of colonic adenomatous polyps,colonic hyperplastic polyps and normal colonic tissues was 90.00%,85.71%,100%,sensitivity was 91.84%,82.76%,100%,specificity was 90.00%,100%,95.92%.The ROC validated the validity of the PCA-LDA method.4.The Spearman correlation analysis showed that there was a positive correlation between fundic gland polyps and colonic adenomatous polyps in the intensity of typical Raman peaks(r=0.9615 P=0.000)and the same as gastric hyperplastic polyps and colonic hyperplastic polyps(r=0.984,P=0.000).5.The abnormal expression of ?-catenin protein in fundic gland polyps(60%)and in colonic adenomatous polyps(53.3%)was not statistically significant(P>0.05).The abnormal expression rate of P-catenin protein in gastric hyperplastic polyps(66.7%)and in colonic hyperplastic polyps(75%)was not significantly different(P>0.05).Conclusion:The near-infrared laser Raman spectroscopy has a certain value in the study of the correlation between different pathological types of gastrointestinal polyps.There were certain physical links between them and its occurrence may be related to the expression of ?-catenin protein,which can provide research basis for the study of the mechanism of simultaneous gastrointestinal polyps.The details as follows:1.Compared with fundic gland polyps and colonic adenomatous polyps,the gastric hyperplastic polyps and colonic hyperplastic polyps had strong mucosal proliferative potential.2.The fundic gastric polyps and colonic adenomatous polyps,gastric hyperplastic polyps and colonic hyperplastic polyps all had a decrease in tyrosine,and ?-catenin protein contains tyrosine,suggesting that the membrane ?-catenin protein expression levels decreased.3.The PCA-LDA method has certain application value in the differential diagnosis of different pathological types of gastrointestinal tissues.4.The fundic gastric polyps and colonic adenomatous polyps,gastric hyperplastic polyps and colonic hyperplasia polyps were linked in substance,and changes in the composition of matter had a certain correlation.5.The occurrence of fundic gland polyps and colonic adenomatous polyps,gastric hyperplastic polyps and colonic hyperplasia polyps were associated with abnormal expression of ?-catenin protein,and may affect the simultaneous occurrence of gastrointestinal polyps through the Wnt/?-catenin pathway.
