Objective: To investigate the distribution of IDH gene mutation and promoter methylation of MGMT gene in different grades of gliomas and whether there is a correlation between them.Methods: From March 2016 to August 2017,67 cases with glioma were removed and diagnosed as brain glioma in the Department of Neurosurgery,the People's Hospital of Xinjiang Uygur Autonomous Region.According to the 2007 edition of the central nervous system and histological classification standard specimens were classified.Sanger sequencing was used to detect whether the IDH gene was mutated.Methylation-specific PCR(MSP)was used to detect whether the MGMT gene promoter was methylated.Mutations in the IDH gene mutation and MGMT gene promoter methylation distribution in different pathological grade gliomas were statistically analyzed using Chi-square test.The correlation between of them was analyzed by Spearman correlation analysis and investigate whether the expression of the them were related to the age and gender of patients with glioma.Results: Among 67 cases of glioma,2 cases of WHO I,14 cases of WHO class ?,18 cases of grade ? and 33 cases of grade ? were found according to the histological classification standard of WHO 2007.1.The results showed that 20 cases(29.9%)detected IDH gene mutation,including 8 cases of WHO ? grade,9 case of ? grade,3 case of ? class.The mutation of IDH gene occurs mostly in WHO?-? grade,and the distribution of it in different grades of gliomas was found to be statistically significant(P<0.05).In patients with age <50 years,the IDH gene utation rate was 36.7% while the rate in patients with age ?50 years old was 11.1%.It as shown that IDH gene mutations were more predisposed to young patients with age 50(P<0.05).The IDH mutation rate was 27.9% in males and 33.3% in females.It seems DH mutations were not correlation with sex(P>0.05).2.The results showed that methylation of MGMT gene promoter occurred in 35 cases(the otal positive rate was 52.2%),among which 1 cases were WHO grade(50%),8 cases of rade ?(57.1%),10 cases of grade ?(55.6%),16 case of grade ?(48.5%).There was o significant difference in the methylation distribution of MGMT gene promoters in ifferent grades of gliomas(P > 0.05)..In patients with age <50 years,the rate ethylation of MGMT gene promoter was 51.1% while the rate in patients with age ?50 ears old was 54.5%.The results showed that there was no correlation between MGMT romoter methylation with age(P>0.05).The methylation rate of MGMT gene promoter as 53.5% in males,while 50% in females.Indicating that there was no correlation etween MGMT promoter methylation with sex also(P>0.05).3.The correlation test between IDH and MGMT showed that 20 cases(29.9%)had IDH ene utation,35 cases(52.2%)detected methylation of MGMT gene promoter,and there was significant correlation between IDH gene mutation and promoter methylation of MGMT ene(P=0.03,r=0.363).conclusion: 1.IDH gene mutations are more likely to occur in patients <50 years old and patients with WHO ?-? grade gliomas.However,there was uncorrelation between IDH gene mutation with gender.2.The MGMT promoter methylation was not related to the patient's gender,age,and pathological grade of the glioma.3.There is a significant correlation between IDH and MGMT gene expression in glioma,and there may be a mutual influence on the gene expression level of glioma.
