Cyclocarya paliurus?Batal.?Iljin-skaja,also called“ready source of money”,“sweet tea tree”because its fruits look like the“copper cash”and the flavor of its leaves.It belongs to the genus Cyclocarya paliurus?Juglangdaceae?deciduous trees,the rare of unique species and medicinal food homologous plants in China,and is also the national key protection endangered species.It is known as the giant panda in the plant kingdom and the third tree of medical field.Modern research showed that,the leaves of Cyclocarya paliurus contained many kind of function components,for instance,sugars,flavonoids,terpenes,proteins,minerals,phenolic acids,vitamins,saponins,etc,among of them,polysaccharides from Cyclocarya paliurus are one of the main functional components,modern research have shown thant the polysaccharides from Cyclocarya paliurus are significant effect on hypoglycemic,lowering blood pressure hypolipidemia,antioxidant,anti-aging,improve immunity and so on,Because of it's extensive biological activity,it has attracted the attention of researchers and has become the focus of research.Currently,the methods for polysaccharides extraction included ultrasonic extraction,microwave assisted extraction,reflux extraction,hot water extraction,ultrasonic enzymatic and ultra pressure extraction.The low extraction rate of polysaccharides from Cyclocarya paliurus has become a bottleneck hindering it's development and utilization,to optimize it's extraction process and improve extraction rate are become very important.In addition,there are few reports on the evaluation of antioxidation and hypoglycemic activity in vitro by the extracorporeal antioxidation model combined with the inhibition ability of an in vitro antioxidant model combined with alpha glucosidase.This topic was based on the hot issues of today's research.The extraction process optimization of the main functional polysaccharides,antioxidant,hypoglycemic in vitro and content dynamic changes,the correlation analysis of polysaccharide content and hypoglycemic activity were studied by modern experimental techniques.It provides a theoretical basis for further research and development and utilization of polysaccharides from Cyclocarya paliurus,the results of the preliminary study are as follows:?1?The extraction process was optimized by response surface method,on the basis of single factor test,the extraction time,material liquid ratio and extraction temperature were taken as the influencing factors.The extraction rate of polysaccharide was the evaluation index.The results showed that the optimum condition was 93?,the extraction time was 2.4 h,the ratio of material to liquid was 1:11,the extraction rate of crude polysaccharide was?4.51±0.25?%.Conclusion:the method is stable and reliable,requiring low equipment condition and convenient control.It can be used for reflux extraction of polysaccharides from Cyclocarya paliurus.?2?To preliminary research the antioxidation and hypoglycemic activity of Cyclocarya paliurus polysaccharides from seven different regions,using the DPPH free radical scavenging capacity?DPPH·?,hydroxyl radical?·OH?,superoxide anion radical?O2-·?and alpha glucosidase inhibitory activity as index.The results showed that the contents of Cyclocarya paliurus polysaccharides from different regions were significant differences?p<0.05?,which Jiangxi Xiushui was the highest?5.85±0.02?%,The Pingyang Township in Rongjiang County of Guizhou was the lowest?3.50±0.10?%.The four indexes have a certain dose effect relationship with the concentration of polysaccharide,that is,as the concentration of polysaccharide increases,the antioxidation and?-glucosidase inhibitory activity will enhance.The IC50 of polysaccharide for four index models was DPPH free radical>glucosidase>·OH free radical>O2-·free radical.?3?To investigate the correlation both the dynamic changes of polysaccharide content and their hypoglycemic activity of different Cyclocarya paliurus in the same habitat.The content of polysaccharides were determined by UV visible spectrophotometry respectively,the activity of inhibiting?-glucosidase was used to evaluate the hypoglycemic effect in vitro,The correlation both the dynamic changes of polysaccharide content and their hypoglycemic activity have been analyzed using SPSS methods.Results:There were significant differences in the content of polysaccharides in different plants under the same habitat,the content of polysaccharides were?1.45%5.84%?,the inhibition rate of polysaccharides from Cyclocarya paliurus on alpha glucosidase was ranged from 73.25%to 97.82%.The positive correlations have been shown significant both the content of polysaccharide and their hypoglycemic activity,the correlation coefficient was 0.661 at the 0.01level.Conclusion:The content of polysaccharide in Cyclocarya paliurus were increased first,then decreased and then increased,the highest content were in June and September.There were significant differences in the content of polysaccharide and their hypoglycemic activityin,It can be used in the deep research and development.?4?To investigate the different processing methods on the content of functional components?flavonoids,polysaccharides,triterpenoids?in Cyclocarya paliurus.Taking Cyclocarya paliurus leaves as material,five post-harvest processing methods?drying at room temperature,direct dry,drying after scalding by boiling water,drying while twisting,drying while twisting after scalding by boiling water?were used determine the three functional components with Ultraviolet spectrophotometric method.Results:The content of total flavonoids was ranged from?17.20±0.36?mg·g-134.10±0.36mg·g-1,the content of polysaccharides was ranged from?38.45±2.21?mg·g-1116.63±2.08mg·g-1,the content of triterpenoids was ranged from?31.45±0.67?mg·g-141.64±0.37mg·g-1.Conclusion:Three functional components were highest in Cyclocarya paliurus leaves by drying while twisting at 80?and 100?.Comprehensive consideration,drying while twisting at 80?was the best post-harvest processing method.
