| Objective:In this paper,the animal model of allergic rhinitis was used as the research object.To research the effects of Press-needle on Dazhui 、 Feishu(double)on allergic rhinitis rats of the symptoms behavioral,pathological and immunological indexes,which provided the experimental basis for the clinical application of Press-needle in the treatment of allergic rhinitis.Methods : Forty Sprague-Dawley(SD)rats of SPF grade were divided into four groups according to the random number table: including model control group,treatment group(Press-needle group),western medicine group(Loratadine group),blank control group,10 rats in each group.The animal model of allergic rhinitis was established by combining intraperitoneal injection of ovalbumin and nasal mucosal stimulation.After setting up the animal model,the treatment group was given a Press-needle treatment,and the Western medicine group had got Loratadine medication.Firstly,we should take an observation and recording of behavioral changes and general situation in rats during the experiment.Unitl the end of treatment,the contents of IgE,IL-4 and IL-2 in serum were measured by RIA,and the histopathologic changes of nasal mucosa were observed under the topical microscope,and statistical analysis of data in each group.Results:1.Symptom scores comparison before and after the treatment: Pretherapy:The rats in the model group were frequent sneezing,scratching and runny nose.Compared with the blank group,the nasal symptom score was significantly higher,the difference was statistically significant,suggesting that the model was successful.After treatment,compared with the Press-needle group、 Loratadine group,the model control group’s symptom score was significantly higher,the difference was statistically significant(P<0.05)2.The results of HE staining of nasal mucosa showed that the nasal mucosa of blank control group rats was intact,smooth and without inflammatory cell infiltration.In the model control group,the epithelium of the nasal mucosa fell off,the inferior connective tissue of the epithelium was necrotic,the glands were proliferated and swollen,and a large number of inflammatory cells were infiltrated.Loratadine group,Press-needle group rats nasal mucosa was obviously improved compared with the model control group,the inflammatory reaction was obviously alleviated,and just a small amount of inflammatory cell infiltration.3.The comparison between the Press-needle group,Loratadine group and model control group had statistically significant difference(all P<0.05)on the expression of IgE and IL-4,and there was little difference of IgE(P>0.05)between Press-needle group and the Loratadine group.4.Compared with Press-needle group,Loratadine group and model group,the differences were statistically significant(all P<0.05)in the serum IL-2 level,but the comparisons between the blank control group with another three groups showed no statistically significant(P<0.05).Conclusion:1.Press-needle therapy has significant effect on relieving nasal symptoms in AR rats.2.Press-needle therapy can effectively reduce the inflammatory reaction of nasal mucosa in AR rats.3.Press-needle therapy can reduce the content of IgE and IL-4 in serum of AR rats,and increase the level of IL-2 AR rats.4.Press-needle may reduce the nasal mucosal inflammation by inhibiting the expression of IgE and IL-4,also through increasing the level of serum IL-2 of AR rats.It makes a big difference for curing the rats.But no significant difference compared with the western medicine group. |
