| 1 Experimental study1.1 Objective To investigate the effects of Yifei Jianpi Granules on the pathological changes,the m RNA and protein expression changes of AQP3 and AQP5 in lung tissue and explore the mechanism of Yifei Jianpi Granules on treating chronic obstructive pulmonary.1.2 Methods Clean SD rats were divided into 7 groups according to the principle of random distribution.(including: blank control group,model group,high dose group of Yifei Jianpi recipe,middle dose group of Yifei Jianpi recipe,low dose group of Yifei Jianpi prescription,ambroxol group and dexamethasone group),There were 16 rats in each group.The rats were 6 weeks and 8 weeks and the weight was 200 ±20 g.The rats were fed normally within 7 days and were tested.On the first day and the 14 day,10% chloral hydrate(0.5ml/100g)was used for abdominal anaesthesia.After successful anesthesia,lipopolysaccharide(LPS)was injected with 200 g g 200 200,and the rats were uniformly rotated,so that lipopolysaccharide was evenly distributed in the lungs.The rats were fumigated twice a day on the 2nd day(except the blank group).The smoke was fumigated in a sealed 1m3 smoking room for 30 minutes in the morning and afternoon.The cigarette was produced by Hongta Group.The tar content was 19 mg and the nicotine was 1.2mg / g.The rats were treated with low-temperature Senna extract for20 days(1 g / ml,1 ml / 100 g body weight,once a day for 30 days).Since then,the model has been built for a total of 49 days.After the model was established,rats were treated by gastric perfusion(except the blank group,the treatment group was divided into three groups: high dose group,middle dose group,low dose group,ambroxol group and dexamethasone group.After 6 weeks of treatment,materials were taken.Lung tissue and bronchus pathological changes were observed by HE staining.AB-PAS staining was used to observe the proliferation of goblet cells and mucin secretion in the bronchi of rats in each group.The expression of MUC5 acm RNA-AQP3 m RNA and APQ5 m RNA in rat lung tissue was detected by RT-PCR.The expression of AQP3 and APQ5 protein were detected by Western-blot and IHC.1.3 Results1.3.1 Effect of Yifei Jianpi on pathological morphology of rats He staining showed that,In comparison between the model group and the blank group,It can be seen that the model group showed cilia shedding in the bronchus.Smooth muscle thickening,inflammatory infiltration,lumen stenosis and other slow obstructive pulmonary pathological changes;Compared with the model group,Yifei Jianpi prescription high,medium and low dosage group,Can alleviate the pathological changes of the bronchi in rats,And with the increase of concentration,the relief is more obvious.The effect of dexamethasone group and ambroxol group was better than that of Yifei Jianpi prescription group.AB-PS staining shows that,In the model group,the proliferation of goblet cells in the bronchus was obvious,And you can see that the stain is getting deeper,In the blank group,the goblet cells did not proliferate obviously.The proliferation of goblet cells was decreased in high dose group,middle dose group,low dose group,ambroxol group and dexamethasone group,The stain shrunk.1.3.2 Effect of Yifei Jianpi Recipe on the content of MUC5 acm RNA,AQP3 m RNA and APQ5 m RNA in the lung tissue and bronchus of rats Compared with the blank group,The content of MUC5 acm RNA in the model group was significantly higher than that in the blank control group,The contents of AQP3 m RNA and APQ5 m RNA were significantly lower than those of the control group(P <0.01).Compared with the model group,the content of MUC5 acm RNA in lung tissue and bronchus of each dose group of Yifei Jianpi recipe decreased to some extent,The content of MUC5 acm RNA in high dose group,dexamethasone group and ambroxol group was significantly decreased(P < 0.01).On the contrary,Compared with the blank group,The contents of AQP3 m RNA and APQ5 m RNA in the model group were significantly lower than those in the blank control group.Compared with the model group,Yifei Jianpi recipe could increase the content of AQP3 m RNA and APQ5 m RNA(P < 0.05).1.3.3 Effect of Yifei Jianpi recipe on the expression of MUC5 ac AQ AQP3 and APQ5 protein in Lung tissue of Rats Compared with the blank group,the expression of MUC5 ac protein in the model group was significantly higher than that in the blank control group.The expression of AQP3 and APQ5 protein was significantly lower than that of control group(P < 0.05).The expression of MUC5 ac protein in lung tissue of rats with high,medium and low dose of Yifei Jianpi recipe was lower than that of model group.The expression of MUC5 ac protein in high dose group,dexamethasone group and ambroxol group was significantly decreased(P < 0.01).On the contrary,compared with the blank group,the expression of AQP3 and APQ5 protein in the lung tissue of the model group was significantly lower than that in the blank control group.Comparison of high,middle and low dose of Yifei Jianpi prescription with model group,The expression of AQP3 and APQ5 protein was increased(P < 0.05).1.3.4 Drug safety Compared with the normal group,no damage of liver and kidney function was found in each dose group and western medicine group.1.4 Conclusion Yifei Jianpi recipe can reduce the expression of MUC5 ac protein and increase the expression of AQP3 and APQ5 protein in lung tissue of rats,so as to alleviate the viscosity of rat airway mucus,make it easier for rats to expel phlegm,and improve the shedding of rat airway cilia.The bronchial smooth muscle thickening and inflammatory cell infiltration were alleviated,and the airway remodeling and pulmonary function decline were delayed. |
