Research purposes: Most prostatitis symptoms are a complex and difficult to cure for patients and doctors.The symptoms are difficult to quantify and effective treatment,which seriously affects patients’ quality of life and physical and mental health.This study aims to investigate the expression of P2X7 R,NLRP3 and IL-1β by constructing a model of autoimmune chronic non-bacterial prostatitis(CNP)in NOD male mice,and to explore the possible role and mechanism of P2X7R-mediated IL-1β in CNP..Methods: Twenty-four mice were randomly divided into four groups,divided into normal control group and CNP model group,ATP+CNP group and BBG+CNP group,with6 rats in each group.After successful anesthesia in mice,in the CNP group,In the ATP+CNP group and the BBG+CNP group,each mouse was injected with a prostatic protein-Freund’s adjuvant emulsion at a subcutaneous point,and then 0.1 ml of DTP vaccine was injected into the thigh muscle of each mouse for two weeks.Then boost the immunization again in the same way.At the beginning of the fourth week after the first immunization,the ATP+CNP group and the BBG+CNP group were intraperitoneally injected with the interventional drug,and the CNP group was injected with the same amount of normal saline once a day for three weeks.Results: The prostate tissue of CNP mouse model was observed to be consistent with the pathological changes of chronic prostatitis under HE staining.The results of immunohistochemistry and Western blot showed that the expression levels of P2X7 R,NLRP3 and IL-1β in CNP model group were all.Compared with the normal control group,the ATP+CNP group had higher P2X7 R,NLRP3 and IL-1β than the CNP model group,and the BBG+CNP group had lower P2X7 R,NLRP3 and IL-1β than the CNP group,and the difference was statistically significant(P< 0.05).Conclusion: P2X7 R mediates the expression level of IL-1β in CNP model and participates in the development and progression of prostatitis.This study provides a new idea for the possible pathogenesis and clinical diagnosis and treatment of chronic prostatitis.
