Objective Mechanism of Shaoyao Decoction in the treatment of ulcerative colitis(UC),based on the mechanical barrier of colonic mucosa and inflammatory cytokines.Methods SD rats were randomly divided into blank control group,model group,mesalazine group and Shaoyao Decoction group.Rat models of ulcerative colitis were replicated by TNBS/ethanol,and each group was treated with normal saline,mesalazine and Shaoyao Decoction.The general condition was recorded.After 7 days of intervention,blood was collected from the abdominal aorta of the rats,and the lesion colon tissue was cut for about 2 cm for histopathology and transmission electron microscopy.Results Compared with the model group,Shaoyao Decoction can alleviate the reduction of body mass,reduce the incidence of diarrhea and blood in the stool,significantly reduce the levels of serum tumor necrosis factor alpha,interleukin-4,interleukin-1β and endotoxin(P<0.05),and reduce the disease activity index and Histopathology score of rats.Under the transmission electron microscope,the tight connection of the model group was obviously damaged.The tight junction destruction of the colonic epithelial tissue of the mesalazine group and the Shaoyao decoction group was lighter than that of the model group.Conclusion Shaoyao Decoction may play a role in the treatment of ulcerative colitis by repairing the tight junction of intestinal epithelial tissue,maintaining mucosal permeability,reducing inflammation and protecting the intestinal mucosal mechanical barrier.
