Objective :Based on the mechanical barrier of the intestinal mucosa and the expression of tight junction protein,the mechanism of Shaoyao Decoction on the treatment of UC rats with damp-heat syndrome is explored,which provides a theoretical basis for the clinical application of Shaoyao Decoction.Methods : 32 SD rats were assigned to 4 groups according to the random number table method,which were marked as blank group,model group,mesalazine group,and Shaoyaotang group.Except for the blank group,the other 3 groups used high-sugar,high-fat spicy diet + TNBS/ethanol method to replicate the damp-heat ulcerative colitis rat model,and used normal saline,mesalazine,and peony decoction to intervene in each group.Record the general conditions,cut out the diseased colon tissue for histopathological and immunohistochemical examination.Results : Compared with the model group,both mesalazine and shaoyao decoction can improve the general condition and dampheat syndrome of UC rats with damp-heat internal accumulation syndrome,and reduce the DAI score and colon histopathological score,suggesting that the treatment group has treatment for the UC rat model with damp-heat internal accumulation syndrome effect.After treatment,immunohistochemistry showed that the content of cyclooxygenase 2(COX2)in colon tissue(P <0.05)was significantly reduced,suggest ed that Shaoyao Decoction can reduce the expression of inflammatory factors in colon tissue and improve the damp heat in UC rats.The immunological indicators of accumulation syndrome,the treatment effect of Shaoyaotang group was significantly better than that of mesalazine group(P <0.05);after treatment,immunohistochemistry showed that the colon tissue of each treatment group was occluded protein-1(Claudin-1)and junction adhesion molecules-1(JAM-1)protein expression increased,and the protein expression level of Shaoyaotang group was similar to that of mesalazine group(P>0.05),higher than that of model group,but lower than that of blank group.Conclusion : Shaoyao Decoction can significantly improve the general condition of UC rats with internal accumulation of dampheat syndrome and the syndrome of internal accumulation of dampheat,reduce the release of inflammatory factors,and increase the colon tissue tight junction associated protein occludin-1(Claudin-1)and junction adhesion molecules-1(JAM-1)expression,thereby promoting the repair of intestinal epithelial tissue tight junctions,protecting the mechanical barrier of the intestinal mucosa to achieve the role of treatment of UC. |
