objective: To investigate the therapeutic effect of Shenlingbaizhu powder on eczema mice with spleen deficiency,to investigate the therapeutic effect of Shenlingbaizhu powder on the recurrence of eczema in mice with spleen deficiency.Methods: 100 healthy male Kunming mice were divided randomly into five groups: normal group,model group,cetirizine group,prednisone group,Shenlingbaizhu powder group.5%,1% DNCB and Senna leaf solution were used to induce allergy and establish mice models of eczema with spleen deficiency.Mice were fed the drug according to the prescribed regimen.After the last dose,ten mice were randomly assigned to each group.The effect of Shenlingbaizhu powder on the eczema mice with spleen deficiency is observed according to the differences in earthickness,ear weight,histopathology in abdominal skin,the count of dermal inflammatory cells and the level of IL-2,IL-4 in serum.After the drug feeding is stopped.reuse 5% DNCB to stimulate the back in mice.The histopathology in the mice back,the count of dermal inflammatory cells and the level of IL-2 and IL-4 in serum were observed by re-induce allergy to establish mice models of eczema with spleen deficiency.Results: 1 Shenlingbaishu powder can reduce the ear thickness difference and ear weight difference in the left and right ears of eczema mice with spleen deficiency Comparing the model group with the normal group,the thickness difference between the left and right ears and the weight difference between the ears of the mice had a significant difference p <0.01.Compared with the model group,cetirizine group,prednisone group and Shenlingbaizhu powder group can effectively reduce the differences in ear thickness and ear weight of the left and right ears of the mice p <0.01.2 Shenlingbaishu powder can increase the level of IL-2 and decrease the level of IL-4 in eczema mice with spleen deficiency The serum levels of IL-2 and IL-4 were significantly different between the model group and the normal group p <0.05.Compare with the model group,the serum levels of IL-2 and IL-4 were significantly increased and decreased in Shenlingbaizhu powder group,cetirizine group and prednisone group p <0.05.3 Shenlingbaizhu powder can reduce the number of infiltration of dermal neutrophils and lymphocytes in eczema mice with spleen deficiency Compared with the normal group,the number of infiltration of dermal neutrophils and lymphocytes in the abdominal tissues of mice in the model group was significantly increased p <0.05.Compared with the model group,the number of infiltration of abdominal derma lneutrophils and lymphocytes was decreased in Shenlingbaizhu powder group,cetirizine group and prednisone group p <0.05.4 Shenlingbaishu powder can improve the pathological tissue of the abdominal skin in eczema mice with spleen deficiency There was no obvious thickening and atrophy of the epidermis in the normal group,and the basal layer was intact.There was no obvious inflammatory cell infiltration around the superficial vessels of the dermis.In the model group,hyperkeratosis,reticular epidermal thickening,spinous hypertrophy,intercellular and intracellular edema were observed.Dermal vasodilation,perivascular inflammatory infiltration.The abdominal skin histopathological structure of the mice in prednisone group and cetirizine group was basically the same.The structure of each layer is basically complete,the hyperkeratosis is not obvious,the spinous layer slightly proliferates.A small number of inflammatory cells surround the vessels of the dermis.Shenlingbaizhu powder group: The epidermis is mildly hyperkeratinized,the spinous layer is mildly proliferative,mild intercellular and intracellular edema,a small number of inflammatory cells infiltrate around the vessels.5 The mice were sensitized again,Shenlingbaizhu powder increased the serum level of IL-2 and decreased the level of IL-4 in mice.The serum levels of IL-2 and IL-4 in the model group were significantly different from those in the normal group p <0.05.Compared with the model group,the serum IL-2 level of the mice was significantly increased and the serum IL-4 level was significantly decreased p <0.05.Compared with the cetirizine group,the serum IL-2 level of the mice was significantly increased and the serum IL-4 level was significantly decreased p <0.05.Compared with the prednisone group,the serum IL-2 level of the mice was significantly increased and the serum IL-4 level was significantly decreased p <0.05.6 The mice were sensitized of infiltration of dermal neutrophils and lymphocytes p <0.05.Shenlingbaizhu powder group was compared with cetirizine group,there are significant differences in the number of infiltration of dermal neutrophils and lymphocytes p <0.05.Shenlingbaizhu powder group was compared with prednisone group,there are significant differences in the number of infiltration of dermal neutrophils and lymphocytes p <0.05.7 The mice were sensitized again,Shenlingbaizhu powder can improve the skin pathological tissue of the mice back The histopathology of the dorsal skin of mice in the prednisone group and the structure of skin tissue on the back of mice in shenlingbaishu powder group is basically complete,hyperkeratosis of epidermis is not obvious,the spinous layer is mildly proliferative,mild intercellular and intracellular edema,a small number of inflammatory cells infiltrate around the vessels.Conclusion:(1)Shenlingbaizhu powder can reduce the inflammatory response of the auricle on the eczema mice with spleen deficiency,increase the level of IL-2,decrease the level of IL-4,and decrease the inflammatory cells of abdominal skin.(2)Shenlingbaizhu powder can reduce the inflammatory response in mice with recurrent eczema,increase the level of IL-2,decrease the level of IL-4,and decrease the inflammatory cells in the skin of the back.The mechanism may be related to its anti-inflammatory effect and improvement of Th1/Th2 imbalance.
