Objective Study of Qi Teng Mai Ning recipe on hematocrit(Hct)、plasma vis-cosity(PV)、endothelin-1(ET-1)、thromboxane B2(TXB2)and 6-keto-prostaglandinF1α(6-Keto-PGF1α)in the plasma of experimental carotid plaque model rats to find the exp-ression mechanism of Qi Teng Mai Ning recipe on carotid plaque formation.Methods 60 middle-aged SPF SD male rats were randomly divided into a blank group,a model group,an atorvastatin group,and Qi teng mai ning recipe group,each group with 15rats.Carotid artery plaque models were prepared in the last three groups.At the 3rd and 12th week of modeling,1-2 rats in the blank group and 2 rats in model group were randomly selected,and the status of model preparation was detected by veterinary B-Ultrasound and HE staining,respectively.After the modeling was completed,the rats in each group were administrated with the corresponding drugs for 4 weeks,and samples were taken.The hematocrit(Hct)and plasma viscosity(PV)expressions were detected by the hemorheology tester;at the same time,an enzyme-linked immunosorbent assay(ELISA)to detect ET-1,TXB2 and 6-Keto-PGF1αin plasma.Results1.Plasma viscosity and Hematocrit Index(1)Compared with the blank group,the plasma viscosity of the rats was significantly increased in the model group(P<0.01).Compared with the model group,the drug viscosity of each drug group was reduced(P<0.05).(2)The expression of hematocrit in rats increased significantly(P<0.01);compared with the model group,the expression of hematocrit in rats decreased(P<0.05).2.ELISA Index(1)Compared with the blank group,the ET-1 in rat plasma was significantly increased in the model group(P<0.01);and the ET-1 in the rat plasma was decreased in each drug group compared with the model group(P<0.05).(2)Compared with the blank group,the TXB2content in the plasma of the model group was significantly increased,and the expression of6-Keto-PGF1αwas significantly decreased(P<0.01);compared with the model group,the atorvastatin group 6-Keto-PGF1αcontent increased,TXB2 expression decreased(P<0.05);Qi Teng Mai Ning Fang group 6-Keto-PGF1αcontent increased,TXB2 expression decreased(P<0.05).Conclusion1.The formation and damage of carotid plaque in aged rats is related to the enhanced expression of Hct、PV、ET-1 and TXB2,and the down-regulation of 6-Keto-PGF1α.2.Qi Teng Mai Ning Recipe can significantly inhibit carotid plaque,improve hemo-rheology and exert vascular protection.Its mechanism may be related to the regulation of Hct、PV、ET-1,TXB2 and 6-Keto-PGF1αexpression disorders.
