Objective:Endothelin (ET), thromboxane A2 (TXA2) and prostacyclin (PGI2) are the three indexes that characterize the function of vascular endothelium. Through the building of type 2 diabetes rats model and the investigation of the effect of Danshen (DS) extraction on the content level of ET and the stable metabolite of PGI2, and thromboxane A2 (TXA2): thromboxane B2 (TXB2) and 6-Ketp-prostaglandin F1a (6-Keto-PGF1α) in blood plasma of diabetes rats, we discuss the mechanism of diabetic angiopathy and the mechanism of the delay formation of diabetic angiopathy through the improvement of the function of vascular endothelium by DS.Method:Healthy male eighty Sprague Dawley (SD) mice weighing 180 ± 10g were selected. Twenty of them were random selected as normal control group, and the rest of them were modeled and injected with 30mg/kg streptozotocin (STZ) by intravascular. Their blood glucose levels were measured after two weeks. The mice of impaired glucose tolerance (IGT) were fed with Hyperglycemia and Hyperlipidemia diet for ten weeks. The forty mice with 2-hour postprandial blood glucose (PBG) above 11.1mmol/L were selected as the successful model. The modeled mice were randomly divided into model group and DS group with each number of twenty. The DS group were intragastricly administered with the DS extraction by 295mg/Kg, while the model group and normal control group were intragastricly administered with saline injection. The feeding was carried out once a day and lasted for two weeks. The changes of body weight and glucose tolerance of the group before and after DS treatment were observed. The contents of ET, TXB2, 6-keto-PGF1α, which is metabolites of TXA2 and PGI2, respectively, were also measured. The experimental results were also statistical analyzed.Results:(1) The glucose endurance with 0, 30, 60 and 120 min of experimental type 2 diabetes mellitus rats was abnormal compared to that of control group, while the DS treatment group showed no evident different compared to that of model group.(2) The ET and TXB2 level in diabetes mellitus (DM) rats of experimental type 2 diabetes rats showed extensively increase compared to the normal control group, while the 6-Keto-PGF1α level was significantly decreased. The content levels of ET and TXB2 in blood plasma of DS treatment rats were significantly decreased, while the content level of 6-Keto-PGF1α was come back to the normal level.Conclusion:(1) The content levels of ET and TXB2 in blood plasma of type 2 diabetes rats can be remarkably decreased, while the content level of 6-keto-PGF1α was increased.
