Objective:To observe the effects of Shenqi Pill "Junior fire supporting vital qi" compatibility on the content of ATP,ADP and AMP and the respiratory chain complex and ATPase activity and ultrastructure in the type 2 diabetes mellitus rats keletal muscle mitochondria with yang deficiency syndrome,explore the modern biological basis of Shenqi Pill in the treatment of type 2 diabetes mellitus with yang deficiency syndrome.Methods:A total of 60 male Sprague-Dawley rats were randomly divided into normal group(12 rats)and model group(48 rats),and the 48 rats were randomly divided into model group,Shenqi Pill group,Bu Yang group and rosiglitazone group after used high sugar and fat feed 3 weeks and small dose streptozotocin and hydrocortisone 2 weeks to induce type 2 diabetes mellitus rats with yang deficiency syndrome.Fasting blood glucose was measured by blood glucose detector;the serum insulin was measured by radioimmunoassay;the blood lipids were measured by automatic biochemical analyzer,the activity of the mitochondrial respiratory chain complex and ATPase in rat skeletal muscle was measured by enzyme-linked immunosorbent assay(ELISA);the contents of ATP,ADP and AMP in skeletal muscle of rats were detected by high performance liquid chromatography(HPLC);the ultrastructure of skeletal muscle was observed under electron microscope.Result:(1)After the success of the model,the rats appeared chills like warm,arch curling up,hi piles,hair erecting or even falling off,slow action,reduced activity,drinking water and food intake reducing,perianal pollution and other Yang deficiency performance in the symptoms and signs.(2)Compared with the normal group,FBG,INS,HOMA-IR,TG,TC,LDL-C weresignificantly increased in the model group,the activity of mitochondrial ATPase,respiratory chain complex and the content of ATP and ADP in mitochondria were significantly decreased and the content of AMP was significantly increased,the difference was statistically significant(P <0.01);compared with the model group,FBG,INS,HOMA-IR,TG,TC and LDL-C were significantly decreased in the Shenqi Pill group and rosiglitazone group,the activity of mitochondrial ATPase,respiratory chain complex and the content of ATP and ADP in mitochondria were significantly increased and the content of AMP was significantly decreased,the difference was statistically significant(P <0.01),but there was no significant difference between the Bu Yang group and the model group(P> 0.05).(3)Compared with the normal group,the skeletal muscle fibers of the model group were disordered,the Z-line and the M-line were blurred and arranged disorderly;the mitochondria were irregular and the volume was smaller,and most of the mitochondria appeared vacuolated and the membrane was dissolved and interconnected;compared with the model group,the skeletal muscle fibers of the Shenqi Pillgroup and the rosiglitazone group were neat,and the Z-line and the M-line were clear and neat,the mitochondrial volume became larger and the vacuoles became less;compared with the Shenqi Pill group,the skeletal muscle of the Bu Yang group was more disordered,the Z-line and the M-line were blurred and the rules were not regular,the mitochondrial structure was incomplete and the vacuolar oceanic change was too small.Conclusions:1.After the treatment of Shenqi Pill,the blood glucose and blood lipid level of type 2 diabetes mellitus rats with yang deficiency syndrome were significantly decreased and the insulin resistance index was improved,showing the drug had good hypoglycemic and lipid-lowering effect.2.After treatment with Shenqi Pill,the activity level of mitochondrial respiratory chain complex Ⅰ,Ⅱ,Ⅲ,Ⅳ and ATPase in skeletal muscle of type 2 diabetes mellitus rats with yang deficiency syndrome was restored;the contentof ATP,ADPincreased,the content of AMP decreased,EC value increased;skeletal muscle mitochondrial ultrastructural pathological changes were improved;thus regulated the skeletal muscle mitochondrial function and body energy metabolism,which may be the modern biology basis of Shenqi Pill "Junior fire supporting vital qi" compatibility can treat type 2 diabetes mellitus with yang deficiency syndrome. |
