| Objective:To study the compatibility mechanism of shenqi pills with "Junior fire supporti ng vital qi".By nuclear magnetic resonance(NMR)to detect kidney bolus of hydrocortisone and chain urea with induced type 2 diabetes mellitus proved the change of serum metabolites in rats,to explore the diagnosis and treatment ki dney pills "Junior fire supporting vital qi" compatibility modern biological basis of kidney Yang deficiency syndrome type 2 diabetes,the interpretation from t he perspective of energy metabolism kidney pills "Junior fire supporting vital q i" compatibility mechanism of action.Methods:A total of 60 male Sprague-Dawley rats were randomly divided into normal gr oup(12 rats)and model group(48 rats),and the 48 rats were randomly divided into model group,Shenqi Pill group,Bu Yang group and rosiglitazone group aft er used high sugar and fat feed 4 weeks and small dose streptozotocin and hy drocortisone 2 weeks to induce type 2 diabetes mellitus rats with yang deficien cy syndrome.Fasting blood glucose was measured by blood glucose Detector.Ea ch treatment group was given the corresponding drug treatment.The tail vein blood was taken to measure fasting blood glucose and blood sugar two hours after the meal with a blood glucose meter.After the end of the experimental i ntervention,the rats were anesthetized,the blood was taken from the inferior v ena cava,and the serum was extracted by centrifugation for nuclear magnetic r esonance detection(NMR).The data was analyzed by R language software and applied by principal component(PCA)and partial least squares method.-Dis criminant analysis(PLS-DA)and other analytical methods to obtain the serum metabolic profile of each group of rats,and draw a load map,then screening and identification of differential metabolites,using multivariate statistical analysi s to compare differential metabolites in the drug-administered group Whether the content of the body changes,in order to speculate on the mechanism of co mpatibility of the prescription of Shenqi Pill "Junior fire supporting vital qi".Result:(1)After the successful preparation of the model,the symptoms and signs of t he rats showed chills,hi together,bow back contracture,body hair sparse,men tal wilting,decreased diet and water intake,decreased activity,slow response,s oft stools,anal A typical yang-like image such as weekly pollution.(2)Analysis of 1H NMR spectrum revealed that 40 kinds of biomarkers w ere present in rat serum.The serum of streptozotocin plus cortisone acetate ma inly contains saccharides,amino acids and derivatives thereof,organic acids,al cohols and the like.According to the change of the content of the substance,i t is inferred that glycolysis,amino acid metabolism,energy metabolism and the like occur in the rat body.Through principal component analysis,each group of samples was within the confidence interval and there were no abnormal sam ples.The normal group,the Shenqi Pill group,the Buyang group,the rosiglita zone group and the model group were separated.Compared with the Buyang g roup and the rosiglitazone group,the Shenqi pill group was closer to the norm al group.(3)Compared with the normal group,the difference of lactic acid,glucose and betaine in the model group was statistically significant(P<0.05).The differenc e between the betaine model group and the kidney gas pill and rosiglitazone g roup was statistically significant(P<0.01),there was no significant difference between the two groups(P>0.05).The difference between the glucose model gr oup and the Shenqi Pill group was statistically significant(P<0.01).The differe nce between the glitazone group was statistically significant(P<0.05),and there was no significant difference compared with the Buyang group(P>0.05).The difference between the lactic acid model group and the Shenqi pill group was different.Statistical significance(P<0.05),the difference was statistically signific ant compared with rosiglitazone group(P<0.01),and there was no significant d ifference compared with Buyang group(P>0.05);The trend of differential meta bolites can be seen from Table 1.Compared with the normal group,the lactic acid and glucose levels are increased and the betaine levels are decreased.Th e kidney qi pills group and the model group are lower in lactic acid and gluc ose levels,while the sugar beet is lower.The alkali level is on the rise.Conclusion:(1)After treatment with shenqi pill,the blood glucose level of rats with type2 diabetes mellitus and kidney-yang deficiency syndrome was significantly redu ced,indicating that the drug had an obvious hypoglycemic effect.(2)After treatment with shenqi pill,the levels of lactic acid,glucose and betai ne in rats with kidney Yang deficiency of type 2 diabetes recovered.Shenqi pi lls maintain the balance of metabolism in vivo by regulating the changes of en ergy metabolites in the body.This may be the biological basis of the compatib ility of shenqi pills with "Junior fire supporting vital qi" in the treatment of ty pe 2 diabetes with kidney Yang deficiency syndrome,and also the mechanism of the compatibility of shenqi pills with "Junior fire supporting vital qi". |
