| Objective: To further study the role of electroacupuncture in preventing atherosclerosis and to construct an intestinal defense system by electroacupuncture,we used a high-fat diet to continuously feed the common carotid artery with balloon injury to simulate atherosclerosis in New Zealand rabbits.In the model,observing the pretreatment of electroacupuncture,the relationship between the morphology of carotid arteries in rabbits,the changes of serum lipids in serum,and the abundance of intestinal flora and community changes in atherosclerosis rabbit models provide experimental evidence for the prevention of atherosclerosis by electroacupuncture.Material and method:Thirty-two male New Zealand white rabbits were randomly divided into control group(n=7),model group(n=19)and electroacupuncture pretreatment group(n=6).The control group was fed with common feed,and the model group was fed with high-fat feed,which did not give exercise time and opportunity every day.After one week of adaptive feeding in the electroacupuncture preconditioning group,electroacupuncture preconditioning was performed.The bilateral internal clearance,bilateral Zusanli and Guanyuan points were selected for electroacupuncture prophylaxis treatment,20 min/time,once daily,acupuncture On the 6th,after taking a rest for 1 week,acupuncture was performed 3 weeks later.Four weeks after high-fat feeding,the model group underwent balloon injury to the common carotid artery and continued high-fat feeding for 4 weeks after surgery.After the completion of the model,one of the control group and the model group was used to determine whether the model was successful or not.HE staining was used to observe the pathological changes of the carotid atherosclerotic plaque in the model rabbits,and the success of the model was identified.Whether or not,the model group was randomly divided into a model group,an electroacupuncture group and a positive drug group,with 6 rats in each group.The electroacupuncture group and the electroacupuncture preconditioning group selected bilateral Neiguan,Zusanli,and Guanyuan points for electrification.Acupuncture treatment,20min/time,once daily.After the treatment is over,blood is collected from the living heart and the common carotid artery tissue and intestinal feces are taken.The pathological changes of atherosclerotic plaques were observed by HE staining.Serum lipid levels were observed in rabbits of each group.High-throughput sequencing methods were used to sequence fecal samples from rabbits with atherosclerosis.Results:1 HE staining resultsCompared with the control group,the arterial endothelium was severely damaged in the model group.Atherosclerotic plaques,inflammatory cell infiltration,and a large number of foam cells were observed under the endothelium.In the electroacupuncture preconditioning group,the blood vessel wall was slightly roughened,and the intima was slightly thickened.The membrane was not smooth and incomplete;compared with the electroacupuncture group,plaque in the electroacupuncture pretreatment group was significantly reduced,indicating that the electroacupuncture preconditioning group was effective in the prevention of atherosclerosis.2 Blood lipid level test resultsCompared with the control group,serum total cholesterol(T-CHO),triglyceride(TG),and low density lipoprotein cholesterol(LDL-L)were significantly higher in the rabbits in the model group,electroacupuncture group,and electroacupuncture preconditioning group(P< 0.01).Compared with the model group,the blood lipids of the electroacupuncture group and the electroacupuncture preconditioning group were decreased,which was statistically significant(P<0.05).There was no significant difference between the electroacupuncture group and the electroacupuncture preconditioning group(P>0.05).3 Intestinal flora sequencing resultsCompared with the control group,the level of gut microbiota in the model group,the abundance of Firmicutes increased,and the abundance of Bacteroidetes decreased,which had statistical significance(P<0.01).Compared with the model group,the abundance of hard endophytic bacteria in the intestinal flora of the rabbits in the electroacupuncture group and the electroacupuncture preconditioning group was decreased,and the abundance of Bacteroidetes was significantly higher(P<0.05).Electroacupuncture preconditioning group Compared with acupuncture group,the abundance of hard endocervula in the electroacupuncture pretreatment group decreased,and the abundance of Bacteroides increased,and the difference was statistically significant(P<0.05).At the family level,the abundances of Ruminococcaceae and unclassified Clostridials were significantly increased in the control group and the model group compared with the model group,while F__S24-7 and Rikenellaceae in Bacteroidetes.As well as the abundance of the Lachnospiraceae of the thick wallworm.There was significant statistical significance(P<0.01).Compared with the model group,the abundance of the rumen family and the unclassified Clostridaceae in the electroacupuncture group and the electroacupuncture preconditioning group decreased significantly,while the bacteroides sp.The abundance of the subjects was statistically significant(P<0.05).Conclusion:1 Electroacupuncture pretreatment of Neiguan,Zusanli and Guanyuan points can protect the atherosclerotic rabbit carotid tissue and prevent the development of atherosclerotic plaque.2 Electroacupuncture pretreatment of Neiguan,Zusanli and Guanyuan points can regulate the levels of T-CHO,TG,LDL-L and HDL-L in the serum of atherosclerosis rabbits,and effectively prevent the changes of blood lipid levels.3 Electroacupuncture Pretreatment of animal models of atherosclerosis can improve the abundance and structure of the flora in animal models,increase the abundance of beneficial bacteria,reduce the abundance of harmful bacteria,and stabilize the steady state of the intestinal flora.It may be one of the mechanisms of acupuncture to prevent atherosclerosis. |
