| Purpose: Selecting guinea pigs acute eczema model as experimental subject,to compare the effects of eczema No.1 on K17,LTB4,LTC4,CysLTR1 and CysLTR2 level and to explore its mechanism of anti-allergic inflammation.Material and method:Eczema is mostly caused by spleen deficiency syndrome.The model of guinea pig spleen deficiency was established by diet and fatigue.The model of acute eczema in guinea pigs was established by Doyora method.The experiment was divided into negative control group,acute eczema group,model group,Chinese medicine low and high dose group of acute eczema of spleen deficiency,Chinese medicine(Huangbai capsule)positive control group,western medicine(loratadine tablets)positive control group.Every day,each one,traditional Chinese medicine high dose group guinea pig gavaged 4ml(including the original medicine 15.5g),traditional Chinese medicine low dose group 4ml / only(including the original medicine 3.875g),Huangbai capsule group 4ml / only(including Huangbai capsule 0.2325g),loratadine tablets group 4ml / only(including loratadine 0.19375mg).The guinea pigs in the negative control group,the acute eczema group and the control group of the acute eczema of the spleen were given equal volume of saline.For 14 days,twice a day.Immunohistochemical method was used to detect the level of K17、CysLTR1 and CysLTR2 in the skin tissues of 7 groups guinea pigs after treatment.The levels of serum LTB4 and LTC4 in 7 groups of guinea pigs were measured by enzyme-linked immunosorbent assay(ELISA).To evaluate the effect of eczema No.1 on allergic inflammation.Result:The treatment of each group could reduce the K17,LTB4,LTC4,CysLTR1 and CysLTR2 levels in varying degrees.Comparing with other treatment groups,the K17 level of traditional Chinese medicine high dose group was the most close to negative control group;the LTB4 level of loratadine group was the most close to negative control group;the traditional Chinese medicine high dose group was the most close to negative control group of LTC4 and CysLTR1 level;while CysLTR2 level of loratadine group was the most close to negative control group.The traditional Chinese medicine high dose group in the levels of K17,LTB4,LTC4,CysLTR1 and CysLTR2 was the most close to negative control group comparing with the low dose group(p<0.05).The levels of K17,LTB4,LTC4,CysLTR1 and CysLTR2 in the traditional Chinese medicine high dose group was the most close to negative control group comparing with the Huangbai capsule group(p<0.05).Conclusion:Eczema 1 could reduce the levels of K17,LTB4,LTC4,CysLTR1 and CysLTR2 from the different degree in acute eczema of guinea pigs,suggesting that eczema 1 had the effect of reducing skin inflammation. |
