Purpose:To explore yam yam extract saponin of painful diabetes peripheral neuropathy in rats serum level of pain caused by material and its mechanism of action,the influence of single taste traditional Chinese medicine in treatment of painful diabetic peripheral neuropathy provides a new means and ideas,at the same time,the optimum choice for clinical diagnosis and treatment of drug provides a deep theoretical foundation.Material and method:Select 120 SPF male Wistar rats,weight 180-200 g,adaptability to feed a week later,randomly selected from 24,it is divided into three groups,each group of 8,respectively as the second week,the fourth week,eighth week normal control group.the remaining 96 rats only for mould making,will be made after the success of the model of rat,were randomly divided into 2 weeks,4 weeks,8 weeks model control group,high dose of group,the yam yam saponins saponins low dose group,prednisone group.Diabetes mellitus was induced by intraperitoneal injection of 2% Streptozotocin(STZ)53 mg/kg(0.1mol /l citrate buffer in PH4.2).Observation for 2 weeks,4 weeks,8 weeks after the mental status and daily activity of rats,blood glucose,and the change of pain threshold,nerve conduction velocity,and using the method of ELISA detecting saponin content of pain onset diabetes peripheral neuropathy pain caused by material in rat serum serotonin,histamine,slow excitation peptide expression.Results:1.The mental state of model group rats,appear more drinks,food,urine with apparent angular,the colour faded lacklustre,nuzzles back arched,slow to wait for a symptom,the treatment group rats are appear afore-mentioned symptoms,but the degree of light in the model group,the yam saponins in high dose group and prednisone group rats symptoms improved relatively obvious.2.The blood glucose level of the model group was significantly higher than that of normal rats(P < 0.01).There was no statistical difference between the blood glucose level and the model group in the treatment group(P > 0.05).3.The motic nerve conduction velocity(MNCV)in the model group was significantly lower than that in the normal group(P < 0.01).The motic nerve conduction velocity(MNCV)was significantly increased in the treatment group compared with the model group(P < 0.01).4.The pain threshold of the model group was significantly higher than that of normal group rats(P < 0.01).In 2 weeks,the pain threshold of diosgenin in high dose group,prednisone group and model group was lower than that in rats(P < 0.05).At 4 weeks,8 weeks,the treatment group compared with model group significantly reduced the pain threshold of rats(P < 0.01),Chinese yam saponins in low dose group and high dose group compared with the pain threshold of rats increased significantly(P < 0.01),prednisone and saponin content of high dose group compared to raise the pain threshold of rats(P < 0.05),prednisone and saponin content low dose group compared with the pain threshold of rats reduced(P < 0.05).5.The levels of pain substances 5-hydroxytryptamine,histamine and bradykinin in rat serum of the normal group were significantly increased(P < 0.01).The serum levels of 5-hydroxytryptamine,histamine and bradykinin in rat serum was significantly decreased compared with the model group(P < 0.01).2 weeks,yam saponins low dose group and saponin content of high dose group compared with the rats serum serotonin,histamine,the levels of slow excitation peptide were significantly increased(P < 0.01),prednisone and saponin content of high dose group compared with the rats serum serotonin levels increased significantly(P < 0.01),prednisone group compared with yam saponins low-dose group rats the levels of histamine and slow excitation peptide in serum were significantly lower(P < 0.01);4 weeks,yam saponins low dose group,prednisone group compared with yam saponins high-dose group rats serum serotonin,histamine,the levels of slow excitation peptide were significantly increased(P < 0.01),prednisone and saponin content low dose group compared with the rats serum serotonin,histamine,the levels of slow excitation peptide were significantly lower(P < 0.01);8 weeks,yam saponins in low dose group,prednisone group and saponin content of histamine in high dose group compared with rat serum,the levels of slow excitation peptide were significantly increased(P < 0.01),prednisone and saponin content low dose group compared with the rats serum serotonin,the levels of slow excitation peptide were significantly increased(P < 0.01).Conclusion:Diosgenin can relieve the pain symptoms of the rats with painful diabetic peripheral neuropathy,and improve the nerve damage in rats with painful diabetic peripheral neuropathy. |
