Clinopodium chinense(Benth.)O.Kuntze is a traditional Chinese herbal medicine.It is the aerial parts of the family Labiatae and known as "Duan Xueliu" in China.Having been used for hundreds of years as a medicine,C.chinense is often used in the treatment of uremia,skin trauma,dysentery,influenza and allergic dermatitis,which has remarkable hernostatic effect.Phytochemical studies have found that there are flavonoids,triterpenoid saponins,diterpenes,volatile oils,fatty acids and many other chemical components in the herb,which have anti-inflammatory,hypoglycemic,antioxidant,anti-tumor and anti-myocardial is chemia pharmacological activitiesCyclocarya paliurus(Batal.)Iljinsk is a unique monotypic plant in China,belonging to the genus Cyclocarya of Juglandaceae.C.paliurus is mainly distributed in the South China's Hubei,Anhui,Fujian and other mountainous areas.The leaf of C.paliurus are often used as raw materials of health food and traditional Chinese medicine for the treatment of diabetes,hypertension and hyperlipidemia,which are widely used and cultivated in China.Phytoch-emistry research shows that the leaves of Cyclocarya paliurus contain flavonoids,triterpen-oids,polysaccharides,amino acids,coumarins and organic acids,which make it have many pharmacological activities,such as anti-inflammatory,anti-oxidation,hypoglycemic and hypolipidemic and so onIn order to fully explore the medicinal value of C.chinense and C.paliurus,the chemical constituents of C.chinense,C.paliurus and their pharmacological effects on diabetic cardiomyopathy were studiedIn this study,the C.chinense and C.paliurus were separated and prepared by silica gel,ODS,Sephadex LH-20 column chromatography,semi-preparative high-performance liquid chromatography and spectroscopic techniques as well as chemical methods.Eight compounds were isolated,purified and identified from 70%ethanol extract of C.chinense and their structures were identified as follows:clinopodiside H(1),nairutin(2),luteolin-7-O-rutinoside(3),didymin(4),clinoposaponin N(5),clinopodiside I(6),clinoposaponin XX(7),buddlejasaponin ?a(8).Among them,compounds 1,5,6 were new compounds.Five compounds were isolated,purified and identified from 70%ethanol extract of C.paliurus,they were identified as follows:Kaempferol(1),medioresinol(2),(+)-terpineol(3),(+)-epiterpineol(4),asiatic acid(5)Using db/db mice as the model,the improvement of diabetic cardiomyopathy of ethanol extract of C.paliurus were studied.The result that ECL can obviously reduce in glucose,TG,TC and MDA levels,and increase the levels of SOD,GSH-Px and CAT of diabetic rats,indicating that ECL has the effect of lowering blood glucose,blood lipid and anti-oxidation ECL improved cardiac function of diabetic rats as evidenced by a remarkable reduction in cTn-I level and LDH,CK,AST activities in the serum.At the same time,the use of HE assay,Masson assay and Western blot experiments found that the ECL reduced the expression of myocardial collagen type I and TGF-?1 as well as proinflammatory cytokines level in myocardial tissue(TNF-?,IL-1? and IFN-?),and increased the P13K,p-AKT phosphorylation levels,indicating that ECL has anti-inflammatory,anti-fibrosis and improved myocardial hypertrophy,and plays an anti-inflammatory effect through the PI3K/AKT signaling pathway;Using the Western blotting to analyze the expression of apoptotic proteins,ECL could reduce Bax,caspase-3 and 9 levels,and increase the Bcl-2 level,indicating its antiapoptotic effect.
