Huperzia serrata(Thunb.)Trev.is a traditional Chinese herbal medicine and widely distributed in South China,Northeast China,and the Yangtze River basin,especially in the areas of damp and mossy ferns.The alkaloid-huperzine A contained in the whole plant of Huperzia serrata has been proved to have a good weakening effect on acetylcholinesterase through animal experiments and clinical validation,and has a significant clinical effect on the treatment of Alzheimer's disease.Due to the clinical side effects of chemical synthesis of Huperzine A,the main source of Huperzine A on the market is isolated from wild resources,and the content of Huperzine A in Huperzia serrata is very low.From 0.006%to 0.100%,people have extensively tapped the wild H.serrata resources,making this species,which is not much of its own species,on the verge of extinction.People try to compensate for the shortage of natural resources through artificial breeding methods,but they have been confined to the laboratory stage.During the course of research,it was found that there are very abundant endophytes in the body of H.serrata.The species and function of these endophytes have become a research hotspot.In this paper,a preliminary bioinformatics analysis of some genomic data of H.serrata was conducted for the first time.A total of 290.44 Gb data was obtained.The genome size was approximately 3.31 Gb,the heterozygosity was approximately 1.50%,and the repeat sequence was approximately 66.15%.The content is about 41.98%,so it is presumed that this species belongs to a type with complex genomic structure,high heterozygosity and reproducibility.Using the method of isolation from tissues,49 strains of endophytic fungi were isolated from different tissue parts of H.serrata at different growth stages.ITS molecular biology was used to identify 5 of them,and a phylogenetic tree was constructed.The five species are Colletotrichum boninense,Colletotrichum fructicola,Hypoxylon sp,Phyllosticta capitalensis and Chaetomium sp.).Cluster analysis was made on the microbial populations in different rhizosphere soils of H.serrata.The differences in bacterial community species between rhizosphere and non-rhizosphere soils were not significant,while the fungal community species in the rhizosphere soil were mainly Ascomycota,Zygomycota and Basidiomycota(Basidiomycota).Among them,Mortierella Coemans,Archaeorhizomyces Archaeorhizomyces sp.,Inocybaceae Inocybaceae sp.,and Aspergillus parasiticus were the dominant flora in the detected rhizosphere soil.According to the relevant data of the sphenernetia serrata genome,the deeper level of the gene can be followed up,and more information on the genetic aspects of H.serrata can be found;the diversity of schizophylla and endophytic fungi and their rhizosphere flora are studied.To lay a foundation for further excavation of the relationship between H.serrata and microbes.
