Objective:To detect the expression of Yes﹣associated protein and DJ-1protein in patients with hepatitis B fibrosis,liver cirrhosis and hepatocellular carcinoma,and to explore YAP,DJ-1 protein and liver function,coagulation function,Correlation of tumor markers,other serological indicators and liver tissue immunological indicators.To analyze the clinical significance of YAP and DJ-1 protein in patients with hepatitis B fibrosis,cirrhosis and hepatocellular carcinoma.Methods:This study is a retrospective study.The liver tissues of 15 patients with liver fibrosis diagnosed with HBV infection and 15 patients with hepatitis B cirrhosis were selected from the Department of Pathology of the First Affiliated Hospital of Guangxi Medical University from September 2016 to February 2019.The cancer tissues of 15 patients with hepatocellular carcinoma were selected;at the same time,15 patients with hepatic hemangioma were selected as the control group(normal liver tissue adjacent to the lesion of hepatic hemangioma was selected).Immunohistochemical staining was used to detect the expression of YAP and DJ-1 protein in each group of tissues.Hematoxylin&eosin staining(H&E)and Sirius red staining were used to observe the liver tissue structure and the degree of fibrosis.The Ishak scores of tissue fibrosis in each group were detected,and the expression of Ariginase-1,Glypican-3,Hepatocyte,CK7,CK19,CD34,Ki67,NM23,P21,P53 and other indicators were detected by immunohistochemistry.Monitor the patient’s liver function,blood coagulation function,blood platelet(PLT),Alpha Fetoprotein(AFP),Protein Induced by Vitamin K Absence or Antagonist-II(PIVKA-Ⅱ),Carbohydrate Antigen 125(CA125),Carbohydrate Antigen 153(CA153),Carbohydrate Antigen 199(CA199),Carcinoembryonic Antigen(CEA)serological indicators,etc.Correlation analysis of the expression levels of YAP and DJ-1 protein with the above serological and immunological indexes respectively.Result:1.Comparison of 4 groups of biochemical indicatorsPIVKA-Ⅱ,TBIL and IBIL,TBA,APTT,CA125,CA153,There was no statistically significant difference between the CEA and CA199 indicators(both P>0.05).There were statistically significant differences in DBIL,AFP,GGT,AST,ALT,PT,TT,and PLT in the 4 groups(all P<0.05).Compared with the normal control group,the hepatitis B fibrosis group,hepatitis B cirrhosis group,and liver cancer group showed an upward trend in indicators GGT,AST,ALT,and PT,and a downward trend in PLT.GGT and PT in fibrosis group,cirrhosis group and liver cancer group were significantly higher than those in control group,and PLT was significantly lower than that in control group(all P<0.05).The AST and ALT levels in the liver cancer group were significantly higher than those in the control group(P<0.05),and the AST levels in the liver cancer group were significantly higher than those in the fibrosis group(P<0.05).2.The expression and quantitative analysis of YAP in 4 groups of organizationsThrough software calculation and analysis,the AO value of YAP in the liver tissue of the control group was(2.85±1.54)×10-3;the AO value of YAP in the fibrosis group was(4.64±4.52)×10-3,and the AO value of YAP in the liver cirrhosis group It is(8.04±11.44)×10-3,and the AO value of YAP in liver cancer group is(1.81±2.13)×10-2,the overall difference is statistically significant(P<0.05).The AO value of YAP was the highest in the liver cancer group,and it was statistically different from the control group,liver fibrosis group,and cirrhosis group(P<0.05).With the severity of the disease,the average optical density(AO)of YAP staining in the control group,liver fibrosis group,cirrhosis group,and liver cancer group gradually increased,and the overall difference was statistically significant(P<0.05).3.Analysis of YAP expression and clinical and pathological indicatorsThe expression of YAP was negatively correlated with the index Hepatocyte(P<0.05);YAP was positively correlated with the indexes AFP,PIVKA-II,AST,ALT,PT(P<0.05).4.Expression and quantitative analysis of DJ-1 in 4 groups of organizationsThe AO value of DJ-1 in the liver tissue of the control group was(1.58±1.52)×10-2;the AO value of DJ-1 in the fibrosis group was(1.44±0.92)×10-2,and the AO of DJ-1 in the cirrhosis group The value was(1.45±2.30)×10-3,and the AO value of DJ-1 in liver cancer group was(3.35±6.46)×10-3,the overall difference was statistically significant(P<0.05).The AO value of DJ-1 was the highest in the control group,and the average value of AO in the 4 groups was:control group>fibrosis group>liver cancer group>cirrhosis group.Compared with the control group,the difference between the cirrhosis group and the liver cancer group was statistically significant(P<0.05);compared with the fibrosis group,the difference between the cirrhosis group and the liver cancer group was statistically significant(P<0.05);the cirrhosis group There was no statistically significant difference between the group and liver cancer group(P>0.05).5.Correlation analysis of DJ-1 expression and clinical indicatorsThe positive AO value of DJ-1 in the 4 groups of tissues was negatively correlated with Ishak score,AFP,AST,ALT,TT of liver fibrosis(P<0.05).Conclusion:1.YAP may be a marker for the deepening of inflammation in patients with hepatitis B fibrosis,cirrhosis,and liver cancer.The increased expression of YAP protein may lead to the progression of liver fibrosis.2.YAP may be a marker for liver cancer screening and early diagnosis.3.DJ-1 protein may be a protein marker that delays the progression of liver fibrosis in patients with hepatitis B.
