Effects Of Modified Huang Qi Jian Zhong Tang On Expressions Of PHD2-mRNA,V-ATPase C-mRNA In Lewis Lung Carcinoma Tumor-bearing Model Mice With Spleen Deficiency Syndrome

Objective To discuss effects of modified Huang Qi Jian Zhong Tang on expressions of PHD2mRNA,V-ATPase c-mRNA in Lewis lung carcinoma tumor-bearing model mice with spleen deficiency syndrome.Method Given by multifactor method(bitter cold purgation,exhausted swimming,irregular fasting),the model mice with spleen deficiency syndrome was established.Each group was dealt with the according plans.Each group was generally observed.The blood was tested by ELISA to detect the expression of the serum gastrin;Then mice were inoculated of Lewis lung carcinoma.The tumor-bearing model mice with spleen deficiency syndrome was established.Mice were administrated with modified Huang Qi Jian Zhong Tang for 11 days.Each group was generally observed.The blood was tested by ELISA to detect the expression of the serum gastrin;Measure the weight of each mouse’s tumor tissue;Tumor tissue was taken for Realtime RT-PCR to detect the expression of PHD2-mRNA and V-ATPase c-mRNA.Result The tumor-bearing model mice with spleen deficiency syndrome was successfully established.The general condition of mice with spleen deficiency syndrome was poor,and the level of serum gastrin were lower(P<0.01).With the treatment of modified Huang Qi Jian Zhong Tang,The general condition of mice with spleen deficiency syndrome was better,and the level of serum gastrin were higher(P<0.01).The tumor-bearing model mice with spleen deficiency syndrome group made the average tumor weight increased(P<0.01),the expression of PHD2-mRNAwas decreased(P<0.01),the expression of V-ATPase c-mRNA was increased(P<0.05).Compared with the tumor-bearing model mice with spleen deficiency syndrome group,the modified Huang Qi Jian Zhong Tang treated tumor-bearing model mice with spleen deficiency syndrome group made the average tumor weight decreased(P<0.05),the expression of PHD2-mRNA was increased(P<0.01),the expression of V-ATPase c-mRNA was of no statistical difference.Conclusion Spleen deficiency factor can make the expression of PHD2-mRNA decreased,make the expression of V-ATPase c-mRNA increased,promote the weight of Lewis lung carcinoma.Modified Huang Qi Jian Zhong Tang can reverse the spleen deficiency syndrome,raise the level of the serum gastrin.Modified Huang Qi Jian Zhong Tang can inhibit tumor growth,which is associated with the increasing expression of PHD2-mRNA.Modified Huang Qi Jian Zhong Tang has no effect on the expression of V-ATPase c-mRNA.
