Objective:Because the etiology of the unexplained recurrent spontaneous abortion is unclear and there is also no clear treating method in the modern medicine,the clinical study was based on the basic theory of TCM and combined with modern medicine.Through the contrast of the decidual tissue expression of interleukin-12 receptor(IL-12 Recipient)and interferon(IFN-γ Recipient)in patients with URSA and in normal premature pregnant women and observing the clinical curative effect of Bushen Huoxue Decoction on the unexplained recurrent spontaneous abortion,further exploring the regulating mechanism of Bushen Huoxue Decoction to IL-12 and IFN-γ cytokine,these will provide a theoretical basis for Bushen Huoxue Decoction treatment to the unexplained recurrent spontaneous abortion.Methods:The experiment will be divided into two parts.In the first part,15 cases of standard patients who suffer URSA pregnancy failure and patients with normal premature pregnancy induced abortion were selected from February 2016 to February 2017,collected the decidual tissue,and examined the expression quantity of L-12Rβ1、IL-12Rβ2、IFN-γ RmRNA within decidual tissue of these two patient groups by using real-time PCR(RT-PCR)detection.The second part:collect 60 standard URSA patients,and then divided them into two groups.The first experimental group of 30 cases were treated by combining Bushen Huoxue Decoction with western medicine flexor progesterone while the control group of 30 cases simply adapt progesterone treatment,and then respectively collected the peripheral blood before and after the two groups treatment.Finally the levels of IL-12 and IFN-γ in both groups were determined by the ELISA method.Results:(1)The expression quantity of IL-12Rβ1 and IL-12Rβ2 within the URSA patients’ decidual tissue were higher than that of patients groups with normal premature pregnancy induced abortion,and it has statistical significance(P<0.05).The expression quantity of IFN-γR in the decidual tissue of the patients with URSA is higher than that of the normal pregnancy abortion group,and this comparison of the two groups was statistically significant(P<0.05).(2)After treatment,the level of IL-12 in the peripheral blood of the URSA patients group is significantly lower than that of the patients with control group after treatment,and the comparison has statistical difference(P<0.01).The level of IFN-y in the peripheral blood of URSA patients after treatment is lower significantly than that of the URSA control group after treatment,and comparing the two groups was statistically significant(P<0.01).(3)The level of progesterone(P),estradiol(E2)and human chorionic gonadotropin(HCG)in the peripheral blood of the URSA patients group were significantly higher than after treatment because of the prescription of kidney invigorating the blood circulation activating and a four weeks flexion progesterone therapy.The two groups are statistically significant(P<0.01);The two groups were statistically significant(P<0.01).The contents of P,E2,and HCG in the peripheral blood of the URSA control groups after a four weeks treatment.Comparison of the two groups were statistically significant(P<0.01).Conclusion:(1)The high levels of IL-12R、IFN-γR in the decidual tissues of URSA patients express the different role of the IL-12、IFN-γ played in patients with URSA and patients in normal pregnancy.It also suggests that IL-12,IFN-γ may participate in the process of URSA.(2)Bushen Huoxue Decoction can lower the expression level of il-12 and IFN-y in the peripheral blood of the URSA patient to improve the clinical efficacy of the patients with recurrent abortion.(3)Bushen Huoxue Decoction content can significantly improve the level of P,E2,and HCG in the peripheral blood of URSA patients to provide the basis for continuing the normal pregnancy.
