| OBJECTIVEAim to establishing a systematic quality evaluation system of Pogostemonis Herba basing on pharmacodynamics indicators,and to select superior-quality production areas by checking the quality of Pogostemonis Herba from different productions in Guangdong province;also objective to determine the drying method of Pogostemonis Herba and to optimize the origin initial processing technology of the medicinal materials,thus providing a scientific basis for the standardization production of Pogostemonis Herba.METHODS1.According to the detection methods and content requirements of the Chinese Pharmacopoeia(2015 edition),all 36 samples batches of Pogostemonis Herba from 7 producinng areas in Guangdong Province have a test on the content of Pharmacopoeia routine inspection items(including impurity content,water content,total ash cotent,acid insoluble ash content and extract content),heavy metals,pesticide residues and organochlorine pesticide,with its content range and quality standard rate also acquired.2.Using steam distillation methord of ChP(2015 edition)to extract the volatile oil of all samples,the oil volume was measured and the yield range was calculated;By adopting the gas chromatography method,the content of pogostone and patchouli alcohol was simultaneously determined with their content range and standard rate of patchouli alcohol also acquired.3.By comprehensively evaluating the five index value,which containing the yield of volatile oil,the total content of pogostone and patchouli alcohol,the Pharmnacopoeia routine inspection items,the content standard rate of five heavy metals content and pesticide residues,the comprehensive quality score was calculated.And the best producing area was selected by comparing the differences of Pogostemonis Herba in each productions.4.The dryness degree,the volatile oil yield,the total content of pogostone and patchouli alcohol,their peak area ratio were used as the index to compare the advantages and disadvantages of three primary processing methods("drying and diaphoretic processing","drying in the shade and diaphoretic processing","drying in the sun and diaphoretic processing")and conclude Parameters of the best drying method by Single-factor test.The optimal primary processing technology of Pogostemon cablin was determined using the orthogonal experiment.The temperature,the single time and the treatment times were used as factors,and L9(34)orthogonal test was designed to optimize and verify the primary processing technology.RESULTS1.The impurity content(%)of all the 36 samples batches of Pogostemonis Herba ranges from 0.09 to 0.60 with the passing rate being 100%;the water content(%)ranges from 5.60 to 13.78,with the passing rate being 100%;the total ash content(%)ranges from 3.72 to 13.56,with the passing rate being 88.89%;the acid insoluble ash content(%)ranges from 0.02 to 3.43,the passing rate is 100%;the extract content(%)ranges from 2.35 to 14.37,with the passing rate being 97.22%;the plum bum element content(Mg/kg)ranges from 0.00~10.99,with the passing rate being 44.44%;the cadmium element content((Mg/kg)ranges from 0.03~0.14,with the passing rate being 0%;the arsenic element content((Mg/kg)ranges from 0.00~1.02,with the passing rate is 97.22%;the mercury element content(Mg/kg)ranges from 0~0.04,with the passing rate being 91.67%;the copper element content(Mg/kg)ranges from 2.68~26.72,with the passing rate being 0%.2.The yield of volatile oil(%)for the 36 samples batches of Pogostemonis Herba is in the range of 0.90~3.10;the content of patchouli alcohol(%)is in the range of 0.13~0.66,the passing rate is 100%;the content of pogostone(%)is in the range of 0.02~0.20.The total content of heavy metals and harmful substances of Pogostemonis Herba from Luoding City is the highest.The Pogostemonis Herba from "Dianbai city" is suprior than others in the total content of patchouli alcohol and pogostone,the Luoding City’s is suprior than others in yield of volatile oil and the content of patchouli.The comprehensive quality score of Pogostemonis Herba from Dianbai city is the highest.3.Each quality index of Pogostemonis Herba show the highest under the "low temperature drying and stuffiness" processing method for Pogostemon cablin.The highest indicator comprehensive score is under 40℃/5 h of drying condition,while the mostoptimized diaphoretic technology is dried at 40 ℃ for 5 h,and diapharetied at temperature of 30℃ for 10 h,4 times of treatment.CONCLUSION1.The Pogostemonis Herba from Guangdong province shows the following trait:the total ash content exceeding,the heavy metals content exceeding and the extact content being insufficient.2.The quality of Pogostemonis Herba from different producing area are different.The test result shows that the medicinal materials quality from Dianbai city is the best,with the highest main medicinal ingredients being and the least heavy metal content that exceeding the standard.3.Among the three investigation factors of the initial processing method of Pogostemonis Herba,the influence of the number of treatments is dominant.Our sdudy of primary processing methods and the optimized processing technology can provide a scientific basis for the improvement of the production technology of Pogostemonis Herba. |
