Objectives: The purpose of the current study was to determine the difference in intestinal microbiota after delivery between healthy fetuses and fetuses with hyperechogenic bowel during the second trimester and the relationship between fetal echogenic bowel and microbiota.Methods: From August 2018 to October 2019,14 healthy fetuses(control group),13 fetuses with echogenic bowel(EB group),and 7 fetuses with echogenic bowel and other abnormalities(C-EB group)were selected.The first meconiums of the newborns before weaning were collected after parturition for high-throughput sequencing of 16 S r DNA.Results:1.After high-throughput sequencing,1267914 effective sequences were generated in all newborn feces samples,and 1222131 high-quality sequences were generated after optimization.A total of 69,222 operation taxonomic units were obtained by clustering all high-quality sequences with 97% similarity.2.The dilution curve of the Goods_coverage index of the three groups of neonatal meconium reached the platform stage,and the sequencing quantity and depth were enough.3.There was no significant difference in Shannon index,Simpson index and Chao 1 index among the three groups.There was no significant difference in αdiversity of intestinal flora among the three groups.4.At the level of phylum,the three groups of newborns formed a community structure mainly composed of Proteus and sclerecta,but the relative abundances of k_bacteria and fusobacteria in the three groups were statistically different(P<0.05).At the level of class,the abundances of γ-Proteus in C-EB group were significantly higher than those in EB group and control group(P<0.05).At the level of order,the relative abundances of Enterobacteriaceae in C-EB group were significantly higher than those in EB group and control group(P<0.05).At the level of family,The relative abundances of Bacillus,Burkholderia and rhizobia in EB group were significantly higher than those in C-EB group and control group(P Two groups(P<0.05).At the genus level,the abundance of Escherichia coli/ Shigella in the control group was significantly higher than that in the EB and C-EB groups(P<0.05),and the abundance of Staphylococcus,Methylbacillus and Campylobacter in the EB group were significantly higher than that in the control and C-EB groups(P<0.05).5.The difference among the control group,EB group and C-EB group were greater than that within the group(R= 0.211)and there were significant difference among the three groups(P = 0.001).Conclusions:1.There was no significant difference in the α diversity and community structure of intestinal flora among the control group,EB group and C-EB group.2.The fetal echogenic bowel in the second trimester is related to the imbalance of intestinal flora.3.The dysbacteriosis of intestinal flora may be one of the factors influencing the fetal echogenic bowel in middle pregnancy. |
