Study On The Medication Rule And Mechanism Of "Fu Ke Cai Zhen" Treating Pregnancy Based On Data Mining Technology

Objective:Pregnancy disease is a key disease in obstetrics and gynecology,which has always been paid attention to by doctors and scholars."Fu Ke Cai Zhen" was written by Feng Huang,a native of Daizhou,Shanxi Province in the Qing Dynasty,which has unique views on pregnancy disease.The author thinks that it is necessary to sort out the prescriptions for pregnancy disease.By using the method of data mining and network pharmacology,this paper studies the prescription law and drug action mechanism of "Fu Ke Cai Zhen" in the treatment of pregnancy disease,and discusses the academic thought and clinical thinking of pre-fetal paper.to guide the modern clinical treatment of pregnancy disease,so that the experience of famous experts can be further passed on.Methods:1.The establishment of the database: sort out all the provisions of the pre-fetal part of "Fu Ke Cai Zhen",extract the relevant elements to establish the prescription database of "Fu Ke Cai Zhen’s pre-fetal part",and process and check the information of the database.The database contains the name of the disease,main symptoms,syndrome elements,prescription name,prescription form,prescription drug composition,drug dose,drug taste menstruation,drug efficacy and so on.2.Data analysis: using SPSS 25.0 software to compare drug use frequency,drug properties,drug taste,drug menstruation,drug.The frequency analysis was carried out in the fields of material efficacy,the number of traditional Chinese medicine prescription,drug dosage,prescription dosage form,main symptoms,syndrome elements and so on.SPSS Modeler 14.1 software was used to analyze the correlation between drugs and symptoms.SPSS 25.0 software was used to carry out systematic cluster analysis of commonly used drugs.The network pharmacology method was used to analyze the mechanism of the basic prescription of "Fu Ke Cai Zhen" in the treatment of recurrent abortion.Results : There are a total of 50 prescriptions for pregnancy in the pre-fetal part of "Fu Ke Cai Zhen",all of which are in accordance with the principles of prescription inclusion.All prescriptions involved 119 traditional Chinese medicines with a total frequency of 362.1.The results of frequency statistics showed that the high frequency drugs were Angelica sinensis,Ligusticum chuanxiong,licorice,Atractylodes macrocephala,Scutellaria baicalensis,Ginseng,Radix Paeoniae Alba and prepared rehmannia root,mainly warm(140 times),pungent taste(176 times),sweet taste(161 times)and bitter taste(127 times).The main meridians were lung meridian(171 times),liver meridian(166times),heart meridian(120 times)and kidney meridian(114 times).The most common drug efficacy is tonifying deficiency(110 times),of which blood tonifying drugs are the most commonly used;the dosage of most drugs is 3-15 g,followed by less than 3g;the number of drugs in the prescription is generally 6-10;the dosage form is mainly decoction;the main symptoms are vaginal bleeding and abdominal pain;the most common disease elements are uterine,followed by lung;blood deficiency is the most common,followed by yin deficiency.2.The results of correlation analysis: the results of drug correlation analysis showed that most of the combinations of two,three and four medicines were the compatibility of high-frequency drugs,and the two drugs with the highest correlation degree were Ligusticum chuanxiong-prepared rehmannia root;the three herbs with the highest correlation were Angelica sinensis-Ligusticum chuanxiong-ginseng;the four herbs with the highest correlation were Angelica sinensis-Ligusticum chuanxiong-prepared rehmannia root-Amomum villosum.Drug-symptom correlation showed that the most commonly used drugs for vaginal bleeding were motherwort,Ligusticum chuanxiong,Angelica sinensis and Donkey-hide gelatin,the drugs with the highest confidence were prepared rehmannia root-Ligusticum chuanxiong,Donkey-hide gelatin-Ligusticum chuanxiong,and licorice for abdominal pain.the drugs with the highest confidence were Ligusticum chuanxiong-Angelica sinensis,Ligusticum chuanxiong-Radix Rehmanniae,Ligusticum chuanxiong-Atractylodes macrocephala,prepared rehmannia root-Angelica sinensis,Atractylodes macrocephala-Angelica sinensis,prepared rehmannia root-Atractylodes macrocephala.3.The results of systematic cluster analysis showed that there were 6 groups of drug combinations: Amomum villosum + prepared rehmannia root + Donkey-hide gelatin +Angelica sinensis+ Ligusticum chuanxiong + Ginseng;Artemisia argyi + Radix Rehmanniae;Radix Scutellariae + Radix Paeoniae Alba;Poria cocos + Radix Ophiopogonis;Platycodon grandiflorum + mulberry white peel;tangerine peel + perilla leaf + pericarpium arecae.4.The results of network pharmacological analysis: after screening,there were 70 basic targets of "Fu Ke Cai Zhen" pregnancy disease,2495 targets related to recurrent abortion and 43 common targets,and the average degree of common target protein interaction was 8.4.9 key targets with high score of cyto Hubba were ESR1,PGR,AR,NR3C1,PPARG,MAPK14,PTGS2,JUN,CASP3.There are two modules in MCODE analysis.Module 1 is related to hormone-mediated signal pathway,DNA template transcription and intracellular receptor signal pathway,and module 2 is related to the regulation of blood pressure,blood circulation and circulatory system.Enrichment analysis involves 632 biological processes,31 cell components,68 molecular functions and 169 signal pathways,such as estrogen signal pathway,p53 signal pathway,PI3K-Akt signal pathway,vascular endothelial growth factor signal pathway and so on.Conclusion: "Fu Ke Cai Zhen" believes that the causes of pregnancy disease are often qi deficiency and blood deficiency,and the deficiency of qi and blood leads to the loss of nourishment in the uterus,so the treatment takes replenishing qi,nourishing blood,relieving pain and calming the fetus as the main method,and the prescription is mainly based on qi,blood,pain and placebo drugs.according to syndrome differentiation plus or minus treatment,the medicine essence prescription is simple.The basic prescription of "Fu Ke Cai Zhen" pregnancy disease is Angelica sinensis-Ligusticum chuanxiong-Radix Rehmanniae-Amomum villosum.The basic prescription plays a role in the treatment of recurrent abortion through hormone regulation,central analgesia,inflammation,immunity and cell survival.It has the characteristics of multi-components,multi-targets and multi-pathways.
