Objective:Based on the theory of "preventive treatment of diseases" of traditional Chinese medicine,this article summarized the medication rules of traditional Chinese medicine diet for diarrhea prevention and treatment by using data mining and network pharmacology,and analyzed the potential mechanism of action of its core prescriptions for the prevention and treatment of diarrhea,aiming to provide scientific basis for the prevention and treatment of diarrhea by traditional Chinese medicine,and provide new ideas for the research and development of new Chinese medicine drugs.Methods:(1)Based on the research methods of literature science and statistics,the literature of traditional Chinese medicine Diet on prevention and treatment of diarrhea in Dictionary of Chinese Medicine Diet(Revised Edition),Complete Record of Traditional Chinese Medicine Diet Prescription and online digital resource database(CNki.com,Wanfang,VIP)was collected as the research object,and the data were standardized collated and the prescription database of medicinal diet was established.Statistical system was used to make statistics on drug frequency,sex and taste normalization,association rules,complex network analysis,etc.On the basis of objective data,the characteristics of drug use of Xiaoxie medicated diet recipe were deeply discussed and analyzed.(2)Active ingredients and targets of drugs in core prescriptions were screened out by TCMSP database,disease targets of diarrhea were obtained from Gene Cards,OMIM,CTD and TTD databases,and intersection targets of drugs and diseases were obtained by Venn platform.The drug-target-disease relationship network was constructed using Cytoscape3.9.0,and the PPI network was constructed using String database.The GO function and KEGG pathway enrichment analysis were conducted on the intersection target using Metascape platform,and the molecular docking simulation verification was conducted using Autodock vina software.And visualize the results.Conclusion:(1)A total of 709 prescriptions of herbal diet for diarrhea were included according to the sodium standard,including 208 kinds of drugs and 237 kinds of food materials.The dosage form of medicated diet is mainly congee,followed by dishes and decoction.The preparation of medicinal food is mainly boiled,followed by decocting,grinding,steaming and stir-frying.The main administration methods were oral administration twice a day,followed by warm and hot taking,fasting eating,meal taking and casual taking.There were 18 drugs with frequency ≥20,and the top 10 drugs were yam(145 times),ginger(122 times),lotus seed(93 times),jujube(62 times),pepper(53 times),Coix seed(53 times),gorgon gorgon(51times),white lentil bean(50 times),poria cocos(49 times)and tangerine peel(47 times).In efficacy analysis,deficiency supplementing drugs were used the most,505 times in total,accounting for 29.78%.Followed by astringent,solution,water infiltration wet,warm,qi,digestion class;The four qi distribution of drugs were mainly warm(787 times,accounting for 46%),followed by flat,cold,hot and cool.The distribution of five tastes was mainly sweet(975 times,accounting for 45%),followed by hot,bitter,sour and salty.The main channel distribution was spleen channel,stomach channel,lung channel and kidney channel.Coix seed,euryale seed-> lotus seed ","Hawthorn,licorice-> Tangerine peel","lotus seed,Coix seed,Euryale seed-> yam" and so on.Through complex network analysis,the core ingredients were yam,euryale,lotus seed,jujube,Coix seed,white lentil and poria cocos.The top ten commonly used foods are japonica rice,sugar,salt,scallion,monosodium glutamate,wine,glutinous rice,brown sugar,soy sauce,vegetarian oil.(2)A total of 82 active ingredients,2329 drug targets,333 disease targets and 65 intersection targets were obtained by network pharmacology analysis of the core medicinal diet formula.The main compounds were quercetin,peroxyergosterol and protoporphyrin.The key targets in PPI network were PIK3R1,PTPN11,STAT3,AKT1,EGFR,IL2,LCK,JAK3,etc.There were 580 overlapping genes enriched molecular functions,4997 biological processes,351 cell components,and 224 KEGG pathways.The key pathways were Th17 cell differentiation,MAPK signaling pathway,calcium signaling pathway,gastric acid secretion,TGF-β signaling pathway,and so on.The antiseptic of Ergosterol-PIK3R1,Ergosterol disinfectant-JAK3,Mauritine The binding energies of D-LCK,Quercetin-PIK3R1,Protoporphyrin-STAT3 and Quercetin-JAK3 were all less than-5.0k J/mol,showing good binding activity. |
