Serum MicroRNA-146a In Breast Invasive Ductal Carcinoma Of The Diagnostic Value And Clinical Index Correlation Analysis

Objective: To compare the expression differences of serum micro RNA-146a(miR-146a)in patients with invasive ductal carcinoma of the breast,fibroadenoma of the breast and healthy women,to evaluate its diagnostic value for invasive ductal carcinoma of the breast,to verify the correlation between the expression of serum miR-146 a in invasive ductal carcinoma of the breast and the clinicopathological characteristics,and to analyze the relationship between the expression of serum miR-146 a and molecular subtypes of invasive ductal carcinoma of the breast.Methods: from 2015 to2016 in our hospital retained in the major disease specimens library serum specimens from30 patients with breast invasive ductal carcinoma,20 patients with breast fibroadenoma,30 cases of healthy women.Use the q RT-PCR technique to detect three sets of peripheral venous blood serum miR-146-a expression level,analysis of its expression differences,application area under the ROC curve evaluation,the sensitivity of the diagnosis of breast invasive ductal carcinoma and specific degrees;Meanwhile,the expression differences of serum miR-146 a among different clinical indicators,molecular indicators and different molecular subtypes of invasive ductal carcinoma of the breast were further analyzed.Results: The expression level of serum miR-146 a in patients with and healthy subjects was similar,and the difference was not statistically significant(P > 0.05).The expression level of serum miR-146 a in patients with invasive ductal carcinoma was significantly higher than that in patients with and healthy subjects.The expression level of serum miR-146 a in patients with invasive ductal carcinoma was statistically different from that in patients with and healthy subjects(P < 0.05),and the area under ROC curve(AUC)was 0.731,(95%CI: 0.604-0.858),the sensitivity,specificity and accuracy were 93.50%,44.40% and 79.70%.There was no significant difference in the expression level of serum miR-146 a among all groups in age,menstrual status,PR,HER-2 expression and tumor size(P > 0.05),but there was significant difference in the expression level of serum miR-146 a among all groups with or without lymph node metastasis and estrogen receptor status(P < 0.05).The expression level of serum miR-146 a was significantly up-regulated with lymph node metastasis.The expression level of serum miR-146 a was significantly up-regulated when estrogen receptor was positive.The expression level of serum miR-146 a was compared among each molecular subtype group,and the expression level of serum miR-146 a was higher in the HER-2 positive HR positive type group,but the difference was not statistically significant compared with each molecular subtype group(P > 0.05).Conclusion: Serum miR-146 a detection has certain diagnostic value in invasive ductal carcinoma of the breast,and can be used as a serological marker for the diagnosis of invasive ductal carcinoma of the breast.The determination of serum miR-146 a expression level is of certain value in evaluating the metastatic invasion ability of invasive ductal carcinoma of the breast.
