Objective: In this study,the rat common carotid artery balloon injury model was used to simulate the clinical restenosis of AS after PTCA,to observe the intervention effect of Huotan Jiedu Tongluo Decoction on the model,and to discuss whether Huotan Jiedu Tongluo Decoction was passed automatically Phage signaling pathway inhibits the formation of atherosclerosis after PTCA.Methods: Thirty-two 5-week-old male(SPF)SD rats were randomly divided into 4 groups,each with 8 rats: sham operation group,model group,traditional Chinese medicine group and western medicine group.After 3 days of adaptive feeding of rats,an AS model after common carotid artery balloon injury was established.Rats in the Chinese medicine group were treated with Huotan Jiedu Tongluo Decoction by gavage,and the Western medicine group was treated with atorvastatin.The other groups were treated with atorvastatin.All were given an equal volume of purified water by gavage,and samples were taken 4 weeks after the operation.1.Using HE staining method to detect the histopathological changes of the injured side of the blood vessel;2.Using immunohistochemistry(IHC)to detect the expression of Beclin-1 in the common carotid artery of each group of rats;3.q PCR detection of each group of rats The m RNA expression levels of autophagy marker molecules Beclin-1,ULK1,Atg12 in common carotid artery tissue.3.Western-blot method was used to detect the expression of ULK1,Atg12 and Atg5 in the common carotid artery of rats in each group.Results: 1.The results of HE staining showed: Compared with the sham operation group,the carotid artery intima of the model group was significantly hyperplasia and the lumen stenosis was more obvious;compared with the model group,the intimal hyperplasia of the traditional Chinese medicine group and the western medicine group were significantly reduced.Lumen stenosis was relieved.2.The IHC test results showed that there were almost no yellow positive expression particles in the sham operation group,and the expression of Beclin-1 was negative;in contrast,the intima of the model group was significantly thickened,and the neointima contained a large amount of The brown-yellow Beclin-1 positive expression particles;compared with the model group,the positive expression of Beclin-1 in the traditional Chinese medicine group and the western medicine group was significantly weakened.The q PCR test results showed that the m RNA expression levels of Beclin-1,ULK1,and Atg12 in the carotid artery tissue of the model group were significantly higher than that of the sham operation group(P<0.01);compared with the model group,the traditional Chinese medicine group The m RNA expression levels of Beclin-1,ULK1,Atg12 in the carotid artery of rats in the Western medicine group were significantly reduced(P<0.01).The western-blot test results showed that: compared with the sham operation group,the protein expression levels of ULK1,Atg12,and Atg5 in the carotid artery tissue of the model group were significantly increased(P<0.01);in contrast,the traditional Chinese medicine group and the western medicine group The protein expression levels of ULK1,Atg12,and Atg5 were significantly reduced(P<0.01).Conclusion: 1.Successfully established the AS model after PTCA in rats through experiments.The study found that its modelling rate is high and the pathological changes are relatively constant,so it can be used to imitate the related experimental research of restenosis after PTCA in humans;2.Phlegm-removing and detoxification Tongluo Yin can significantly inhibit the intimal hyperplasia after carotid balloon injury,and improve the formation of carotid atherosclerosis in rats;3.The autophagy signal pathway is overexpressed in the AS model after PTCA in rats,and the phlegm and detoxification are eliminated.Tongluo Yin can effectively inhibit the excessive activation of the autophagy pathway,which provides a new treatment strategy and molecular biological basis for the future treatment of AS after PTCA with traditional Chinese medicine.
