| Objective:To investigate the effect of moxa stick moxibustion on the expression of 11β-hydroxy steroid dehydrogenase type 1,11β-HSD1 mRNA and protein in amygdala and hippocampus of rats with corticosterone-yang deficiency.Possible Mechanism of Suspension Moxibustion in Treating Deficiency of Kidney Yang.Methods:Thirty-four healthy Sprague-Dawley male rats were divided into a blank control group(A n = 6)and a modeling group(n = 28).All rats in the modeling group were injected with hydrocortisone at a dose of 0.03 mg / g · bw.(Alternative injection of both lower limbs),the modeling was performed for 14 days.After the modeling was completed,4dead rats were excluded,and the remaining 24 rats were randomly divided into a model group(B n = 6),a suspension moxibustion group(C n = 6),a blocking agent group(D n = 6),Suspended moxibustion + blocker group(E n = 6).Rats in groups A and B were bundled daily without any other treatment.Rats in group C were given acupuncture moxibustion "Shenshu" and "Guanyuan" points.Group D was alternately injected with gallic acid on both hind limbs.The rats were given moxa stick moxibustion and gampere acid in the same way as group C and group D.Each group was treated once a day,one course on the 7th,and the second course was continued after 2 days of rest,and the treatment was performed 14 times.After treatment,all rats were sacrificed and tissue samples were collected.Adrenocorticotropic hormone(ACTH),corticotropin-releasing hormone(CRH),and corticosterone(CORT)in rat serum were observed by ELISA,and observed by Western blot technology.The expression of 11β-HSD1 protein in hippocampus and amygdala of rat brain was observed.The expression of 11β-HSD1 mRNA in hippocampus and amygdala of rat was observed by RT-PCR.Results:1.Changes in behavior and weight of rats: before modeling,all rats are healthy,have a good diet,strong mobility,soft and shiny hair,red ears,enlightened eyes,strong and strong resistance,and returned to the cage.Scrambled and beat each other.There was no statistically significant difference in body weight between the groups(P> 0.05),indicating that the experiment was comparable;after modeling,the rats appeared thin,rough hair,obvious hair loss,curled up in the corners,dull eyes,pale ears,Cold limbs,cold tails,poor mobility,and other signs of kidney yang deficiency.The weight value of group A increased significantly compared with the model group(P<0.01).After treatment intervention in each group,the rats’ food intake and water intake increased,their activity increased significantly,their body hair gradually returned to smooth and smooth,stool gradually formed,urine output decreased,their limbs became cold,their tails cooled,and their hair removal improved,which proved that the modeling was successful After treatment,the symptoms of kidney yang deficiency improved.2.Detection of rat serum indicators: Compared with group A,the serum CORT,ACTH,and CRH of group B rats were significantly reduced(P<0.01),indicating that the modeling was successful.Compared with group B,the serum levels of CORT,ACTH,and CRH in rats in each treatment group increased to varying degrees,of which C and E groups were the most significant(P<0.01),and ACTH and CRH in group D had extremely significant differences(P<0.01),there was a significant difference in CORT(P<0.05).This shows that the moxa stick moxibustion can increase the levels of serum CORT,ACTH and CRH in different degrees.3.Expression of 11 β-HSD1 mRNA in hippocampus and amygdala:(1)Compared with group A,the expression of 11β-HSD1 mRNA in hippocampus and amygdala of group B was significantly reduced(P <0.01),indicating that 11β-HSD1 mRNA content decreased.(2)Compared with group B,the expression of 11β-HSD1 mRNA in hippocampus of each treatment group increased significantly(P<0.01),and the amygdala increased significantly in each group(P<0.05).This shows that the moxa stick moxibustion can effectively increase the expression of 11β-HSD1 mRNA in hippocampus and amygdala of kidney-yang deficiency rats.(3)There was no statistical difference between the treatment groups(P> 0.05).4.Expression of 11β-HSD1 protein in hippocampus and amygdala:(1)Compared with group A,the expression of 11β-HSD1 protein in hippocampus and amygdala of group B was significantly decreased(P <0.01),indicating that 11β-HSD1 protein content decreased.(2)Compared with group B,the expression of 11β-HSD1 protein in hippocampus and amygdala of rats in each treatment group increased to varying degrees,and both were statistically significant(P <0.05),indicating that moxa suspension moxibustion can effectively increase kidney yang deficiency 11β-HSD1 protein expression in rat hippocampus and amygdala.(3)There was no statistical difference between the treatment groups(P> 0.05).in conclusion: 1.Suspension moxibustion of Shenshu and Guanyuan points can increase the levels of serum ACTH,CRH and CORT,relieve the inhibition of HPA axis,and improve the symptoms of kidney-yang deficiency syndrome.2.Moxa stick moxibustion up-regulates the expression of 11β-HSD mRNA and protein in hippocampus and amygdala,which may be one of the mechanisms of moxa stick improvement moxibustion for improving kidney yang deficiency.Conclusion:1.Suspension moxibustion of Shenshu and Guanyuan points can increase the levels of serum ACTH,CRH and CORT,relieve the inhibition of HPA axis,and improve the symptoms of kidney-yang deficiency syndrome.2.Moxa stick moxibustion up-regulates the expression of 11β-HSD mRNA and protein in hippocampus and amygdala,which may be one of the mechanisms that moxa stick improves moxibustion of kidney-yang deficiency. |
